摘  要:试验旨在分离鉴定出鸽乳中产角蛋白酶微生物,并研究其产酶特性。通过筛选培养基分离出目标菌株,观察菌落形态,测定角蛋白酶活力,提取菌株基因组DNA并扩增其16S rDNA序列,测序结果经BLAST比对和相似性分析,并构建系统发育树。此外,还探究了菌株在不同时间点的猪皮降解率和角蛋白酶活性变化规律,以及在不同底物条件下的角蛋白酶活性大小。结果:分离出3个产角蛋白酶菌株,1株为大肠杆菌(Escherichia coli),2株为枯草芽胞杆菌(Bacillus subtilis);上述菌株均具有较好的产酶能力,且48 h后对猪皮的降解率均超过60%;3种菌株所产角蛋白酶的活性随着时间增长而逐渐升高;3种菌株均对羊皮存在较高的降解作用,其次为猪皮和鸡皮。试验结果为鸽营养物质消化机理和产角蛋白酶微生物的研究与应用奠定基础。The aim of this experiment was to isolate and identify keratinase-producing microorganisms from pigeon milk and to determine the characteristics of the enzymes they produce.The strains were isolated by screening media and their colony morphology was observed.The activities of keratinase were determined.Then,the genomic DNA of the strains was extracted,and their 16S rDNA sequences were amplified.The sequencing results were compared by blast and similarity analyses,and a phylogenetic tree was constructed.In addition,the changing patterns of pig skin degradation rate and keratinase activities of the strains at different time points were explored.Meanwhile,the activities of keratinase under different substrates were also investigated.The results showed that three keratinase-producing strains were isolated:one of Escherichia coli and two of Bacillus subtilis.All the above strains had good enzyme production capacity.The degradation rate of the above strains on pig skin exceeded 60%at 48 h of the experiment.The activities of keratinase increased gradually as the experimental time went on.In addition,all the three strains had a higher degradation effect on sheep skin,followed by pig skin and chicken skin.The results of this ex⁃periment provided a basis for research on nutrient digestion and on application of keratinase-producing microorganisms in pigeon breeding.
