水包油型灵芝多糖纳米乳佐剂的制备及乳化方法探究

摘  要:研发一种水包油型灵芝多糖纳米乳(GLP-NE)佐剂,初步应用于非洲猪瘟病毒P72重组抗原制剂研究,以提升P72免疫制剂稳定性和有效性。本研究利用伪三元相图法确定最佳组方,采用剪切搅拌、超声2种乳化方法筛选制备GLP-P72纳米乳的工艺参数,并检验其安全性、物理性状及免疫效果。结果表明,灵芝多糖纳米乳佐剂为水包油型且符合纳米乳佐剂要求,粒径为(167.4±0.1)nm,黏度为4.2 s·(0.4 mL)^(-1),1 500 r·min^(-1)剪切搅拌20 min可制备出较为稳定的纳米乳。该佐剂经济有效,乳化方法简单稳定,可使GLP-P72纳米乳具有较好的稳定性和免疫效果,并对中药成分纳米乳佐剂的研发和制备提供借鉴。In this study,an oil-in-water Ganoderma lucidum polysaccharide nanoemulsion(GLP-NE)adjuvant was developed to improve the stability and effectiveness of African swine fever virus(ASF)P72 recombinant antigen preparation.The optimum formulation was deter⁃mined by pseudo-ternary phase diagram,and the technological parameters of the preparation of GLP-P72 nano-emulsion were screened by shear stirring and ultrasonic emulsification.Finally,the safety,physical properties and immune effects of the preparation were tested.The re⁃sults showed that Ganoderma lucidum polysaccharide nano-emulsion adjuvant was of an oil-in-water type and met the requirements of nanoemulsion adjuvant.The particle size was(167.4±0.1)nm,the viscosity was 4.2 s·0.4 mL^(-1),and the stable nano-emulsion could be prepared by shearing and stirring 1500 r·min^(-1) for 20 min.The adjuvant prepared here was economical and effective,and the emulsification method was simple and stable,which enhanced the stability and effectiveness of GLP-P72 nano-emulsion,and could serve as reference for research on and development of nano-emulsion adjuvant as an element of traditional Chinese medicine.
