摘  要:为了解不同剂量蒲虎颗粒对脂多糖(lipopolysaccharide,LPS)诱导的小鼠急性肺炎治疗效果,试验将60只小鼠(雌雄各半)随机分为6组,分别为蒲虎颗粒低剂量组(每天按体重2 g/kg)、中剂量组(每天按体重4 g/kg)、高剂量组(每天按体重8 g/kg)、蒲地蓝消炎颗粒对照组(每天按体重3.2 g/kg)、模型对照组和空白对照组,每组10只,雌雄各半。给药组按体重0.1 mL/10 g灌胃给药,每天2次(间隔6 h),连续给药3 d。第1次给药结束后麻醉小鼠,给药组及模型对照组将LPS(50μL,按体重2.5 mg/kg)滴入小鼠咽后壁造模,空白对照组给予等体积的磷酸缓冲盐溶液(phosphate buffer saline,PBS)。给药期间每天观察小鼠的精神与行为状况;试验结束后,处死小鼠,采集气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)和肺脏组织,对肺脏组织进行肺炎症状评分并称重,制作石蜡切片采用苏木精-伊红(hematoxylin-eosin,H.E.)染色观察小鼠肺脏组织病理学变化;用细胞计数板计数BALF中白细胞数;用试剂盒分别采用BCA法和ELISA法检测总蛋白和肺脏组织匀浆中肿瘤坏死因子-α(tumor necrosis factor,TNF-α)质量浓度。结果表明:蒲虎颗粒给药后第2天可改善小鼠的精神与行为状况;与空白对照组相比,模型对照组肺脏组织出现大片实变灶、肺泡间隔增厚、炎性细胞渗出等症状,给药组小鼠肺脏组织病变均有不同程度的减轻。与模型对照组相比,蒲虎颗粒中、高剂量组及蒲地蓝消炎颗粒对照组的肺炎症状评分均极显著降低(P<0.01);蒲虎颗粒中、高剂量组的肺脏重量极显著降低(P<0.01);蒲虎颗粒中、高剂量组及蒲地蓝消炎颗粒对照组BALF中白细胞数、总蛋白质量浓度及肺脏组织匀浆中TNF-α质量浓度均极显著降低(P<0.01)。说明中剂量蒲虎颗粒就能减轻LPS诱导的肺脏组织损伤,抑制肺水肿、蛋白质渗出及相关炎症因子释放,对LPS诱导的急性肺炎模型具有较好的治In order to understand the therapeutic effects of different doses of Pohu granules on lipopolysaccharide(LPS)-induced acute pneumonia in mice,in this experiment,sixty mice were randomly divided into six groups:Puhu granules low-dose group(2 g/kg per day according to body weight),medium-dose group(4 g/kg per day according to body weight),high-dose group(8 g/kg per day according to body weight),Pudilan Xiaoyan granules control group(3.2 g/kg per day according to body weight),model control group,and blank control group,with 10 mice in each group,half male and half female.The drug groups were given by gavage 0.1 mL/10 g body weight twice a day(6 h apart)for 3 days.After the first dose was completed,the mice were anesthetized,and LPS(50μL,2.5 mg/kg body weight)was instilled into the posteripharyngeal wall of mice in the drug groups and the model control group,and the blank control group was given an equal volume of phosphate buffer saline(PBS).The mental and behavioral conditions of mice were observed daily during administration.After the experiment,mice were sacrificed,bronchoalveolar lavage fluid(BALF)and lung tissue were collected;pneumonia symptoms were scored and weighed on lung tissue;paraffin sections were made for hematoxylin-eosin(H.E.)staining to observe the histopathological changes of the mice in lung tissue.The number of leukocytes in BALF was counted with a cytometer;BCA and ELISA methods were used to detect the total protein and the mass concentration tumor necrosis factor-α(TNF-α)in the lung tissue homogenate using the kits.The results showed that the mental and behavioral conditions of mice could be improved on the second day after administration of Puhu granules.Compared with the blank control group,the lung tissue in the model control group had symptoms such as consolidation foci,thickening of alveolar septum,and exudation of inflammatory cells,and the lung tissue lesions of mice in the drug groups were reduced to varying degrees.Compared with the model control group,the symptoms of pneumonia in
