摘  要:为了探究脂肪酸合成代谢相关基因在梅花鹿不同组织中的相对表达量及其差异性表达规律,试验选取过氧化物酶体增殖物激活受体家族成员PPARγ、激素敏感脂肪酶(HSL)、脂肪酸合成酶(FAS)三个与脂肪酸合成代谢相关的基因,分别选取雌性、雄性梅花鹿的心脏(乳头肌)、肝脏(左叶)、肺脏(右肺上叶)、肌肉(背最长肌、肋间肌、臀大肌、股四头肌)和皮下脂肪8种组织样品,采用实时荧光定量PCR技术,检测PPARγ、FAS和HSL基因的相对表达量并探究其差异性表达规律。结果表明:PPARγ、FAS、HSL基因在梅花鹿各组织中均有不同程度的表达,且表达量存在组织、性别间差异;三者在雌性、雄性梅花鹿皮下脂肪组织中的表达量均极显著高于其他组织。三种基因在梅花鹿不同组织中表达量顺序分别为:PPARγ基因,在雄性个体中,皮下脂肪>心脏>肺脏>肝脏>肋间肌>背最长肌>股四头肌>臀大肌;在雌性个体中,皮下脂肪>肺脏>肝脏>心脏>肋间肌>背最长肌>臀大肌>股四头肌。FAS基因,在雄性个体中,皮下脂肪>肝脏>肺脏>心脏>肋间肌>背最长肌>股四头肌>臀大肌;在雌性个体中,皮下脂肪>肺脏>肝脏>肋间肌>心脏>背最长肌>臀大肌>股四头肌。HSL基因,在雄性个体中,皮下脂肪>肝脏>肺脏>背最长肌>肋间肌>臀大肌>股四头肌>心脏;在雌性个体中,皮下脂肪>肝脏>肺脏>肋间肌>背最长肌>股四头肌>臀大肌>心脏。说明PPARγ、FAS和HSL基因在梅花鹿不同组织中的相对表达量与梅花鹿性别、脂肪含量有关,该结果将为进一步选育肉质优良的梅花鹿提供新的理论参考。In order to investigate the expression levels and differential expression patterns of the fatty-acid-synthesis-metabolism-related genes in sika deer,three genes of peroxisome proliferator-activated receptor PPARγ,hormone sensitive lipase(HSL)and fatty acid synthase(FAS)were analyzed in eight types of tissues of both sexes,including heart(papillary muscle),liver(left lobe),lung(upper lobe of right lung),muscle(longissimus dorsi,intercostal,gluteus maximus,quadriceps femoris)and subcutaneous fat,by the means of real-time fluorescence quantitative PCR.The results showed:The different expression levels of PPARγ,FAS and HSL genes were observed in eight types of tissues of sika deer,showing gender variance,and the highest expression of three genes was observed in subcutaneous adipose tissues of both sexes.The orders of three gene expression levels in different tissues of both sexes were as follows:For PPARγgenes,subcutaneous fat>heart>lung>liver>interostal muscle>longest dorsal muscle>leg muscle>gluteal muscle in males,and subcutaneous fat>lung>liver>heart>intercostal muscle>longest dorsal muscle>gluteal muscle>leg muscle in females,respectively;For FAS genes,subcutaneous fat>liver>lung>heart>intercostal muscle>longest dorsal muscle>leg muscle>gluteal muscle in males,and subcutaneous fat>lung>liver>intercostal muscle>heart>longest dorsal muscle>gluteal muscle>leg muscle in females,respectively;For HSL genes,subcutaneous fat>liver>lung>longest dorsal muscle>intercostal muscle>gluteal muscle>leg muscle>heart in males,and subcutaneous fat>liver>lung>intercostal muscle>longest dorsal muscle>leg muscle>gluteal muscle>heart in females,respectively.It could be concluded that the expressions of PPARγ,FAS,and HSL genes were tissue-specific and influenced by the sex and body adipose deposits in sika deer,and identification of differential expression of PPARγ,FAS,and HSL genes in sika deer would facilitate the breeding of high-quality sika deer.
