辽宁绒山羊低温保存精液稀释液配方筛选研究

摘　　要：为了研究辽宁绒山羊精液稀释液对精液低温保存效果的影响,前期试验研制了10个配方,用于绒山羊精液保存,测定了低温保存不同时间的精子活力;根据试验数据,初选了低温保存时间最长、活力最高的精液稀释液配方。以此配方为基础,通过调整三羟甲基氨基甲烷(Tris)、乙二胺四乙酸(EDTA)、柠檬酸(1H_(2)O)的添加剂量,形成了4组精液稀释液配方,进一步测定了精液低温保存不同时间的精子活力,从中确定了效果最优的精液稀释液最终配方:三羟甲基氨基甲烷(Tris) 2.42 g、柠檬酸(1H_(2)O) 1.34 g、乙二胺四乙酸(EDTA) 0.20 g、葡萄糖1.00 g、新鲜卵黄20.0 mL、超纯水80.0 mL、青霉素1.0×10^(5) IU、链霉素1.0×10^(5) IU。以最终配方配制辽宁绒山羊精液稀释液,对绒山羊原精进行稀释,低温保存4 d后进行辽宁绒山羊人工输精,统计了母羊的受胎率。结果表明:以最终配方对辽宁绒山羊精液进行稀释,低温保存到第4天时的精子活力为(66.78±5.12)%,用于配种后母羊受胎率达到91.3%。综上所述,筛选出的辽宁绒山羊精液稀释液配方保存和使用效果良好,满足了辽宁绒山羊人工授精工作中对精液的长距离、长时间运输和保存的需求,可以在辽宁绒山羊人工授精工作中推广应用。In order to study the effect of diluents on cryopreservation of semen in Liaoning cashmere goat,10 formulations were developed for cryopreservation,and sperm motility of semen at different cryopreservation time was measured.According to the test data,the formula of semen diluent with the longest cryopreservation time and the highest vitality was chosen for the experiment.Four groups of semen diluent formulations were formed by adjusting the added doses of Tris,EDTA and citric acid(1H 2O),and the sperm motility of semen at different low temperature storage times was further investigated,from which the final formula of semen diluent with the best effect was determined:Tris 2.42 g,citric acid(1H 2O)1.34 g,EDTA 0.20 g,glucose 1.00 g,fresh yolk 20.0 mL,ultra-pure water 80.0 mL,penicillin 1.0×105 IU,streptomycin 1.0×105 IU.The semen diluent was prepared with the final formula and used to dilute the fresh semen of Liaoning cashmere goat,and then stored at low temperature for 4 days for artificial insemination breeding of Liaoning cashmere goat.The results showed that the sperm motility was(66.78±5.12)%,and the estrus conception rate reached 91.3%.In summary,the determined formula of semen diluent in Liaoning cashmere goat showed positive effects of preservation at low temperature and application,which could meet the requirements of long-distance and long-term transportation and preservation of semen for the artificial insemination in Liaoning cashmere goat,showing prospects of its extension in the production of Liaoning cashmere goat.
