凋亡细胞疗法在器官移植排斥反应中的作用机制

摘　　要：器官移植是目前器官衰竭的最好治疗方法,但移植患者需终生口服免疫抑制剂控制移植排斥。免疫耐受是机体对自身成分或部分外来抗原的免疫无应答现象,探索其机制有助于器官移植排斥的防治。凋亡的淋巴细胞可抑制细胞或体液介导的免疫反应,凋亡细胞处理可诱导和维持外周免疫耐受,减轻慢性炎症性疾病进展。在器官移植中,供体凋亡细胞处理可减少部分免疫抑制剂用量,其机制可能与凋亡细胞通过下调免疫细胞的抗原呈递能力诱导免疫耐受现象有关。基于目前的研究结果,凋亡细胞疗法有望对临床诱导器官移植免疫耐受提供新的思路。Organ transplantation is currently the best way to treat various organ failures,but the transplant patients depend on life-time immunosuppressants to control transplant rejection.Transplantation tolerance is the phenomenon of the body′s immune unresponsiveness to its own components or some foreign antigens,and it is helpful to exploring its mechanism for preventing organ transplantation.Apoptotic lymphocytes can suppress cellular or humoral mediated immune responses.Apoptotic cells processing can induce and maintain peripheral immune tolerance,and alleviate the progression of chronic inflammatory diseases.In organ transplantation,the treatment of donor apoptotic cells can reduce the dosage of some immunosuppressants,the related mechanism of which might be associated with down-regulating the antigen-presenting ability of immune cells by apoptotic cells.Based on current research results,the apoptotic cell-based therapy is expected to provide new ideas for the clinical induction of immune tolerance in organ transplantation.
