摘 要:川崎病是多因素诱导的一种小儿急性血管炎性疾病,可累及中小动脉,引起冠状动脉病变。研究认为川崎病的发生发展与表观遗传学改变密切相关,DNA甲基化是表观遗传学研究的主要内容之一,在目的基因转录过程中发挥关键作用。DNA甲基化在川崎病患儿维持机体正常细胞结构和功能中占据重要地位,这为通过改变目的基因甲基化状态进而改善急性期川崎病炎性损伤、免疫调节紊乱、冠状动脉病变、静脉注射免疫球蛋白治疗耐药和寻找新的防治靶点等提供了理论依据。未来,DNA甲基化检测在川崎病患儿的临床诊断、治疗、疗效评价及预后中具有良好的应用前景。Kawasaki disease is a multifactorial induced acute vascular inflammatory disease in pediatric patients,which can involve small and medium-sized arteries and cause coronary artery lesions.Studies suggest that the development of Kawasaki disease is closely related to epigenetic alterations,and DNA methylation is one of the major components of epigenetic studies which plays a key role in the transcription of target genes.DNA methylation plays an important role in the maintenance of normal cellular structure and function in children with Kawasaki disease,which provides a theoretical basis to improve the inflammatory damage,immune regulation disorders,coronary artery lesions,drug resistance to intravenous immunoglobulin therapy and search new targets for prevention and treatment by changing the methylation status of target genes.In the future,DNA methylation testing has a promising application prospect in the clinical diagnosis,treatment,efficacy evaluation and prognosis of children with Kawasaki disease.
