摘  要:目的研究长链非编码RNA(lncRNA)母系表达基因3(MEG3)对宫颈癌细胞自噬的影响。方法将人宫颈癌细胞系HeLa分为两组,对照组转染空载质粒的HeLa细胞,实验组使用病毒转染方式对HeLa细胞系进行过表达MEG3基因。使用实时荧光定量聚合酶链反应技术测定HeLa细胞中MEG3基因的信使RNA(mRNA)表达水平,然后采用CCK8法检测细胞增殖情况,流式细胞术检测细胞凋亡情况。利用Western blot技术检测HeLa细胞中自噬标记蛋白[微管相关蛋白1轻链3(LC3)-Ⅱ/LC3-Ⅰ]的表达水平。结果实验组lncRNA MEG3 mRNA表达水平显著高于对照组(750.59±7.30比1.01±0.04)(t=324.911,P<0.001)。不同时点间HeLa细胞增殖率的主效应差异有统计学意义(P<0.01),不考虑测量时间两组间的主效应差异有统计学意义(P<0.01),组间和时点间存在交互作用;与对照组相比,实验组不同时点间HeLa细胞的细胞增殖能力明显降低(P<0.05)。实验组HeLa细胞总凋亡率明显高于对照组[(32.43±0.50)%比(19.67±0.05)%](t=36.032,P<0.001)。两组LC3-Ⅰ蛋白表达比较差异无统计学意义(P>0.05),实验组LC3-Ⅱ蛋白表达、LC3-Ⅱ/LC3-Ⅰ比值低于对照组(P<0.01)。结论lncRNA MEG3可以抑制宫颈癌自噬和增殖活性,促进肿瘤细胞的凋亡,有望成为宫颈癌治疗的靶点。Objective To investigate the effect of maternally expressed gene 3(MEG3)in long non-coding RNA(lncRNA)on autophagy of cervical cancer cells.Methods The human cervical cancer cell line HeLa was divided into two groups:the control group was transfected with HeLa cells with empty plasmid,and the experimental group used the viral transfection of the HeLa cell line to overexpress MEG 3 gene.The messenger RNA(mRNA)expression level of MEG3 gene in Hela cells was measured by RT-qPCR technology,and the cell proliferation was detected by CCK8 method and apoptosis was detected by flow cytometry.The Western blot technique was then used to detect the protein expression level of autophagy marker protein[microtubule-associated protein 1 light chain 3(LC3)-Ⅱ/LC3-Ⅰ]in Hela cells.Results The lncRNA MEG3 mRNA expression level in the experimental group was significantly higher than that in the control group(750.59±7.30 vs 1.01±0.04)(t=324.911,P<0.001).The main effect difference of HeLa cell proliferation rate between different points was statistically significant(P<0.01);regardless of measurement time,the main effect difference between the groups was significant(P<0.01),and there was an interaction between groups and time points;HeLa cells proliferation capability at different time points were significantly reduced compared with the control group(P<0.05).The total apoptotic rate of HeLa cells in the experimental group was significantly higher than that in the control group[(32.43±0.50)%vs(19.67±0.05)%](t=36.032,P<0.001).There was no significant difference in LC3-Ⅰprotein expression between the two groups(P>0.05),and the expression of LC3-Ⅱprotein and LC3-Ⅱ/LC3-Ⅰof the experimental group was lower than those of the control group(P<0.01).Conclusion LncRNA MEG3 can inhibit autophagy and proliferative activity of cervical cancer,promote apoptosis of tumor cells,and is expected to become a target for cervical cancer treatment.
