摘 要:目的观察针刺强刺激对类痛经大鼠的镇痛效应及二酰甘油(DAG)/蛋白激酶C(PKC)信号通路的影响。方法40只雌性SD大鼠采用随机数字表法分为空白组、模型组、针刺弱刺激组和针刺强刺激组,每组10只,适应性饲养7 d后,采用苯甲酸雌二醇联合缩宫素注射法建立类痛经模型。其中,空白组与模型组不予针刺干预,针刺弱刺激组予Φ0.20 mm×1.5 mm揿针治疗,针刺强刺激组给予Φ0.25 mm×40 mm一次性针灸针进行深刺并行平补平泻手法,两组分别留针20 min后,观察大鼠的扭体反应。比较4组的扭体潜伏期和扭体评分,子宫平滑肌细胞内Ca 2+平均荧光强度(MFI),血清和子宫组织中的DAG水平,钙调蛋白依赖性蛋白激酶Ⅱ(CaMKⅡ)水平和PKCγ蛋白表达的变化。结果模型组的扭体潜伏期短于空白组[(251±90)s比(505±76)s]、扭体评分高于空白组[(34.2±25.0)分比(5.9±2.1)分](P<0.01);针刺弱刺激组、针刺强刺激组的扭体潜伏期长于模型组[(354±109)s、(513±76)s比(251±90)s](P<0.05或P<0.01),针刺强刺激组的扭体评分低于模型组[(9.9±6.6)分比(34.2±25.0)分](P<0.01);针刺强刺激组的扭体潜伏期长于针刺弱刺激组(P<0.01)、扭体评分低于针刺弱刺激组[(9.9±6.6)分比(23.1±11.9)分](P<0.05)。模型组大鼠子宫平滑肌细胞内Ca 2+MFI显著高于空白组(P<0.01),针刺强刺激组和针刺弱刺激组大鼠子宫平滑肌细胞内Ca 2+MFI均明显低于模型组(578±134、588±288比1099±872)(P<0.05)。模型组血清和子宫组织中的DAG水平均明显高于空白组(P<0.01),针刺强刺激组和针刺弱刺激组血清和子宫组织中的DAG水平均明显低于模型组[(8.5±2.6)μg/L、(9.0±3.6)μg/L比(14.4±3.1)μg/L,(6.5±3.4)μg/L、(7.3±2.6)μg/L比(13.4±4.2)μg/L](P<0.01)。模型组的CaMKⅡ水平显著低于空白组(P<0.01),PKCγ蛋白表达水平高于空白组(P<0.01);针刺强刺激组的CaMKⅡ水平显著高于模型组(0.24±0.07比0.17±0.07)(P<0.01),PKCγ蛋�Objective To observe the analgesic effect of strong-stimulation acupuncture on dysmenorrhea model rats and its influence on diacylglycerol(DAG)/protein kinase C(PKC)signaling pathway.Methods Forty female SD female rats were randomly divided into a blank group,a model group,a weak-stimulation acupuncture group and a strong-stimulation acupuncture group,with 10 rats in each.After 7 days of adaptive feeding,dysmenorrhea model was established by estradiol benzoate combined with oxytocin injection.No intervention was performed in the blank group and the model groups.The weak-stimulation group was treated withΦ0.20 mm×1.5 mm press needle.The strong-stimulation group was treated with deep acupuncture and moderate reinforcing and reducing method withΦ0.25 mm×40 mm disposable acupuncture needle.After 20 minutes of needle retention in both groups,the writhing response of all rats were observed.The latency and number of twists in four groups,mean fluorescence intensity(MFI)of Ca 2+in uterine smooth muscle cells,DAG levels in serum and uterine tissue,calmodulin-dependent protein kinaseⅡ(CaMKⅡ)levels,and PKCγprotein expression level changes of the four groups were compared.Results The latent period of writhing in the model group was obviously shorter than in the blank group[(251±90)s vs(505±76)s](P<0.01),while the writhing score was significantly higher than that in the blank group(34.2±25.0 vs 5.9±2.1)(P<0.01).The latent period of writhing in the weak-stimulation acupuncture group and strong-stimulation acupuncture group were obviously longer than that in the model group[(354±109)s,(513±76)s vs(251±90)s](P<0.05 or P<0.01),and the writhing score in the strong-stimulation acupuncture group was significantly lower than that in the model group(9.9±6.6 vs 34.2±25.0)(P<0.01).The latent period of writhing in the strong-stimulation acupuncture group was obviously longer than that in the weak-stimulation acupuncture group(P<0.01),and the writhing score was significantly lower than that in the weak-stimulation acupun
