摘 要:肾脏纤维化是几乎所有肾脏疾病进展至终末期肾病的共同病理改变,严重威胁人类健康。Rho/Rho相关卷曲螺旋蛋白激酶(ROCK)信号通路是生物体内广泛存在的经典信号通路,可通过调节骨架蛋白的聚合状态影响细胞收缩、黏附、迁移等生物学行为。Rho/ROCK信号通路的异常激活参与肾脏纤维化的发生发展,有望成为临床治疗肾脏纤维化的有效靶点。因此,深入研究Rho/ROCK信号通路介导肾脏纤维化发生发展的机制,可以为慢性肾脏病的临床诊治和新药研发提供思路。Renal fibrosis is a common pathological change in almost all kidney diseases that progress to end-stage renal disease,posing a great threat and challenge to human health.The Rho/Rho-associated coiled-coil containing protein kinase(ROCK)signaling pathway is a classical signaling pathway widely existing in organisms,which can affect a variety of biological behaviors such as cell contraction,adhesion,and migration by regulating the polymerization of cytoskeleton.The aberrant activation of Rho/ROCK signaling pathway is involved in the occurrence and development of renal fibrosis,which is expected to become an effective target for the clinical treatment of renal fibrosis.Therefore,in-depth studies of the mechanism of the Rho/ROCK signaling pathway in mediating the occurrence and development of renal fibrosis can provide ideas for the clinical diagnosis and treatment of chronic kidney disease and the development of new drugs.
