摘 要:贵州省某养鸡场两栋鸡舍中的蛋鸡疑似发生了鸡传染性喉气管炎病毒感染,为确定该病病因,引物按已发表的TK基因引物序列进行合成,从发病鸡采集的病料中扩增出1259 bp大小的片段;将阳性病料组织处理后接种LMH细胞,盲传3代后发现LMH细胞出现CPE现象,收集其病毒液对其进行PCR鉴定、生长曲线的测定以及动物回归试验,确定分离到一株ILTV毒株,并将其命名为GZNY202112株。为了进一步了解其遗传特征,根据GenBank发表的鸡传染性喉气管炎病毒TK基因、gD基因设计了特异性引物,使用DNAstar软件绘制遗传进化树。TK以及gD遗传进化分析结果显示该毒株与澳大利亚原产疫苗株CSW-1、V1-99亲缘关系最近,GZNY202112株可能与ILT疫苗株有关。In order to determine the cause of infectious laryngotracheitis virus infection,the laying hens in two chicken houses of a chicken farm in Guizhou province were suspected to have been infected with infectious laryngotracheitis virus.Primers were synthesized according to the published primers sequence of TK gene and 1259 bp fragments were amplified from the infected chicken materials;LMH cells were inoculated with positive samples after tissue treatment,CPE phenomenon was found in LMH cells after 3 generations of blind transmission,the virus was collected and identified by PCR,growth curve determination,and animal regression experiments to determine an ILTV strain,which was named GZNY202112 strain.To learn more about its genetic characteristics,specific primers were designed according to TK gene and gD gene of infectious laryngotracheitis virus of chicken published by Gen Bank,and DNAstar software was used to draw the genetic evolution tree.Evolution analysis showed of TK and gD genetic showed that this strain was closely related to the Australian vaccine strains CSW-1 and V1-99,and GZNY202112 might be related to the ILT vaccine strain.
