摘  要:为了了解森林革蜱Septin-2基因的结构、表达特征及生物学功能,通过对Septin-2基因进行克隆、原核表达及生物信息学分析,利用RT-qPCR方法检测该基因在森林革蜱不同发育时期以及不同组织间的表达丰度。结果显示,本实验室培养的森林革蜱庆阳株Septin-2基因中最大编码区为1281 bp,可编码大小为426 aa的蛋白,主要功能区基因大小为868 bp,与森林革蜱陕西株(KAH7952853,XP_037569269)的同一性最高,高达100%;森林革蜱Septin-2在卵巢中的表达丰度最高,且蜱饱血后的表达量明显高于饥饿状态的蜱,Septin-2基因能够表达37 ku的重组蛋白,并能被家兔抗森林革蜱血清识别。上述研究表明,Septin-2基因在蜱饱血及卵巢的发育过程中均表达。In order to understand the structure,expression characteristics and biological function of Septin-2 gene in Dermacentor silvarum,the possible functions of Septin-2 was resolved by cloning,prokaryotic expression and bioinformatics analysis,and the RT-q PCR was used to detect the expression abundance of the Septin-2 in different developmental stages and different tissues of D.silvarum.The results showed that the maximum coding domain size of Septin-2 gene was 1281 bp,and encoding426 amino acids size protein,and the main functional region gene was 868 bp in size.The identity of the gene was 100%with that of shaanxi strain(KAH7952853,XP_037569269).The abundance of Septin-2 expression was highest in the ovary,and was significantly higher after engorgement than that in un-engorgement ticks.The recombinant Septin-2 protein was 37 ku in molecular weight,and can be identified with rabbit anti-D.silvarum serum.This study indicated that Septin-2 was expressed in ticks engorgement and ovarian development.
