摘  要:试验旨在探究芳香烃受体(AhR)活化对免疫应激仔猪结肠菌群及短链脂肪酸含量的影响。选择21日龄健康状态良好、体重相近的断奶仔猪24头,随机分为4组,每组6个重复,每个重复1头猪。对照组(CON)仔猪饲喂基础日粮,LPS应激组(LPS)仔猪屠宰前腹腔注射100μg/kg BW脂多糖(LPS)。FICZ组(FICZ)仔猪腹腔注射250μg/kg 6-甲酰基吲哚并[3,2-B]咔唑(FICZ)+屠宰前注射LPS,CDN组(CDN)仔猪腹腔注射6 mg/kg豆蔻明(CDN)+屠宰前注射LPS。试验期15 d。结果显示,与LPS组相比,FICZ组丙酸、丁酸和戊酸含量显著增加(P<0.05);CDN组仔猪结肠Chao1、Observed species指数显著增加(P<0.05),戊酸含量增加。与LPS组相比,在门水平上,FICZ和CDN组仔猪结肠蓝藻门相对丰度明显增加,厚壁菌门相对丰度明显降低;在属水平上,FICZ和CDN组仔猪结肠Anaerotruncus、梭状芽孢杆菌属、阿利斯佩斯菌属、盐单胞菌属、气微菌属、Sediminispirochaeta、帕拉普菌属、粪球菌属、副萨特菌属、Intestinimonas、互养菌属、柯林斯氏菌属、Barnesiella和Odoribacter相对丰度明显提高。研究表明,AhR活化可调控仔猪结肠肠道菌群组成及短链脂肪酸的合成,缓解LPS诱导的肠道微生物屏障损伤。The study aimed to investigate the effect of AhR activation on intestinal flora and short-chain fatty acid content in immune-stressed piglets.A total of 24 weaned piglets(Duroc×Landrace×Yorkshire)with good health and similar weight at 21 d were randomly assigned into four groups with six replicates in each group and one pig in each replicate.Piglets in control group(CON)were fed with a basal diet,and piglets in LPS stress group(LPS)were intraperitoneally injected with 100μg/kg BW LPS before slaughter.Piglets in FICZ group(FICZ)were intraperitoneally injected with 250μg/kg FICZ+LPS before slaughter,and piglets in CDN group(CDN)were intraperitoneally injected with 6 mg/kg CDN+LPS before slaughter.The experiment lasted for 15 d.The results showed that compared with LPS group,the content of propionic acid,butyric acid and valeric acid of FICZ group increased significantly(P<0.05),the Chao1 index and Observed species index in CDN group increased significantly(P<0.05),and valeric acid increased.Compared with LPS group,at the phylum level,the relative abundance of Cyanobacteriain in FICZ group and CDN group increased significantly and the relative abundance of Firmicutes decreased significantly.At the genus level,the relative abundance of Anaerotruncus,Cloacibacillus,Alistipes,Halomanas,Aeromicrobium,Sediminispirochaeta,Paraprevotella,Faecalicoccus,Parasutterella,Intestinimonas,Synergistes,Collinsella,Barnesiell and Odoribacter was significantly increased in FICZ group and CDN group.The study indicates that AhR activation can regulate the composition of colonic microbiota and the synthesis of short-chain fatty acids in piglets,and alleviate LPS-induced intestinal microbial barrier damage.
