雾化吸入威视派克对提高大鼠肺组织分辨率的实验研究

摘  要:目的:通过雾化吸入威视派克(VSP)对比剂提高肺组织分辨率的方式,对大鼠肺部进行显像,以实现大鼠气道显像的目的。方法:选择SPF级雄性SD大鼠5只,体重90~100 g,随机分为对照组1只、实验组4只:对照组雾化吸入生理盐水2 ml,实验组分别雾化吸入剂量为0.1 ml/kg VSP、0.3 ml/kg VSP、0.5 ml/kg VSP、0.7 ml/kg VSP与生理盐水混悬液共2 ml;所有大鼠在麻醉后即时、雾化吸入生理盐水或VSP后0、30、60、90、120、150、180 min分别进行肺部CT扫描,通过比较肺组织CT值变化以及对比CT三维气道重建影像,了解雾化吸入VSP对肺部显像情况的影响。结果:对照组肺组织CT值及气道显像情况在雾化吸入生理盐水前后无明显变化;实验组随着雾化吸入VSP剂量的增加,其肺部组织CT值随之升高,于120 min达峰值,且其气道三维重建影像在雾化吸入VSP后显像更佳。结论:雾化吸入VSP提高大鼠肺组织分辨率及气道成像的方法是可行的:雾化吸入剂量为0.5 ml/kg VSP对于提高大鼠肺组织分辨率效果最佳,且最佳显像时间为120 min。Objective:To visualize the airway and improve the resolution of lungs in rats with aerosol inhalation of Visipaque(VSP).Methods:Five SPF male Sprague-Dawley rats,weighing 90 to 100 g,were randomly divided into control group(n=1)and experimental group(n=4).The control group inhaled normal saline(NS)2 ml,and the experimental group inhaled 2 ml suspension with 0.1 ml/kg,0.3 ml/kg,0.5 ml/kg,0.7 ml/kg VSP and NS,respectively.Each rat was scanned with CT for 8 times(after anesthesia,after inhaling NS or suspension for 0,30,60,90,120,150,180 min).After that,compare CT value changes of lung tissue and reconstruction of airway imaging.Results:There were no significant changes in CT value of lung tissue or airway visualization in the control group after aerosol inhalation NS or not.However,for the experimental group,CT value of lung tissue increased with VSP concentration increasing,reaching peak at 120 min.And the airway reconstruction images would be better after inhaling aerosolized VSP.Conclusion:It is feasible to improve the resolution of lungs and visualize airway with aerosol inhalation of VSP.For our study,0.5 ml/kg was the optimal dose for improving the resolution of lungs,and best image could be gotten after inhaling 120 min.
