摘  要:目的探讨二陈汤抑制高脂饲养肥胖小鼠的炎症和肝脏脂肪变性作用和机制。方法将45只C57BL/6雄性小鼠随机分为对照组15只、高脂组30只,分别给予普通饲料和高脂饲料喂养,连续6周。高脂组小鼠体质量高于对照组20%判定为肥胖小鼠造模成功。随后将高脂组随机分为模型组和二陈汤组各15只,二陈汤组每天给予含生药浓度0.555 g/mL二陈汤药液20 mL/(kg·d)灌胃,对照组和模型组给予等体积无菌生理盐水灌胃,连续4周。然后处死小鼠,HE染色观察肝脏组织形态学改变;ELISA检测小鼠血清中甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)和脂多糖(LPS)含量;ELISA检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)以及脂多糖结合蛋白(LBP)的含量;qPCR和Western blot法分别检测小鼠肝脏组织Toll样受体-4(TLR4)和髓样分化因子88(MyD88)mRNA相对表达水平及蛋白表达量。结果与对照组比较,模型组肝小叶结构不清晰,肝脏细胞发生脂肪变性,小鼠体质量,血清TG、TC、LDL-C、TNF-α、IL-6、LBP、LPS均明显升高(P<0.05),TLR4 mRNA相对表达水平明显降低(P<0.05),MyD88 mRNA相对表达水平明显升高(P<0.05),TLR4和MyD88蛋白表达量均明显升高(P<0.05);与模型组比较,二陈汤组肝脏细胞病理学形态改变明显减轻,血清TG、TC、LDL-C、TNF-α、IL-6、LBP、LPS均明显降低(P<0.05),TLR4、MyD88 mRNA相对表达水平均明显降低(P<0.05),TLR4、MyD88蛋白表达量均明显降低(P<0.05)。结论二陈汤可降低LPS抑制TLR4、MyD88通路,改善肥胖小鼠的炎症和肝脏脂肪变性。Objective:To investigate the effect and mechanism of Erchen Decoction on inflammation and liver steatosis in high-fat diet-induced obese mice.Methods:A total of forty-five C57BL/6 male mice were randomly divided into the control group(15 mice)and the high-fat group(30 mice),fed with the normal and high-fat diet respectively.After 6 weeks,mice of the high-fat group were divided into the model group and the Erchen Decoction(ED)group,with 15 mice in each group.The ED group was given 0.02 L/(kg·d)of Erchen Decoction by gavage while the control and model groups were given sterile saline by gavage for 4 weeks,then all mice were executed.The levels of triacylglycerol(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)and lipopolysaccharide(LPS)were determined by ELISA.HE staining was used to observe the histomorphological changes in the liver.ELISA for serum levels of tumour necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and lipopolysaccharide binding protein(LBP).The expression of toll-like receptor-4(TLR4)and myeloid differentiation factor 88(Myd88)of liver in mRNA and protein levels were measured by qPCR and Western blot.Results:Compared with the control group,the structure of lobules of liver was not clear,and hepatic cells had steatosis in mice of the model group,which had significantly higher body mass,serum TG,TC,LDL-C,TNF-α,IL-6,LBP and LPS(P<0.05),significant lower level of TLR4 mRNA(P<0.05)and higher level of Myd88 mRNA(P<0.05),however significant higher level of TLR4 and Myd88 protein(P<0.05).Compared with the model group,pathological morphological changes of liver cells significantly reduced,serum TG,TC,LDL-C,TNF-α,IL-6,LBP,and LPS significantly decreased in the ED group(P<0.05),and expression of TLR4 and Myd88 significantly reduced at mRNA and protein levels(P<0.05).Conclusion:The result of this study suggested Erchen Decoction could reduce inflammation and liver steatosis in obese mice,which might due to decrease of LPS and inhibition of TL
