亚麻木酚素干预下ApoE基因敲除小鼠肝脏转录组分析

摘  要:利用转录组测序技术研究了亚麻木酚素干预下ApoE-/-(Apolipoprotein E,ApoE)小鼠肝脏组织中差异表达基因及相关信号通路,为明确亚麻木酚素缓解高脂血症的分子机制奠定基础。结果表明,亚麻木酚素组和模型组之间,共有372个差异表达基因,其中包括234个显著上调基因,138个显著下调基因。这些关键的差异表达基因包括Hsd3b5、Acot1、Elovl3等与脂质代谢相关的基因。GO(Gene ontology,GO)富集分析发现差异表达基因主要富集在脂肪酸代谢等生物过程、细胞外间隙等细胞组分以及单加氧酶活性等分子功能。KEGG(Kyoto encyclopedia of genes and genomes,KEGG)信号通路富集分析表明差异表达基因在PPAR(Peroxisome proliferation activated receptors,PPAR)信号通路被富集。PPAR信号通路的多个基因的表达被激活,如Fabp、Cyp、Acsl、Ehhadh、Fads、Acaa、Scp、Pltp等,推测亚麻木酚素是通过PPAR信号通路调节ApoE-/-小鼠血脂水平的。Transcriptome sequencing technology was used to study the differentially expressed genes and related signaling pathways in the liver of ApoE-/-mice under the intervention of flax lignans,so as to lay the foundation for clarifying the molecular mechanism of flax lignans in alleviating hyperlipidemia.The results showed that there were 372 differentially expressed genes between the flax lignans group and the model group,including 234 significantly up-regulated genes and 138 significantly down-regulated genes.The key differentially expressed genes included Hsd3b5,Acot1,Elovl3 and other genes related to lipid metabolism.GO enrichment analysis showed that the differentially expressed genes were mainly enriched in biological processes such as fatty acid metabolism,cellular components such as extracellular space,and molecular functions such as monooxygenase activity.KEGG signaling pathway enrichment analysis showed that the differentially expressed genes were enriched in PPAR signaling pathway.The expression of multiple genes of PPAR signaling pathway was activated,including Fabp,Cyp,Acsl,Ehhadh,Fads,Acaa,Scp,Pltp,etc.It was speculated that flax lignans regulated the blood lipid level of ApoE-/-mice through PPAR signaling pathway.
