摘 要:目的比较银质封闭式玻璃化冷冻系统(SSCVS)、开放式冷冻薄膜(Cryotop)和传统封闭式麦管(CPS)对第3天分裂期胚胎的冷冻保存效果。方法选择2017年1月至2018年10月在四川大学华西第二医院行体外受精-胚胎移植(IVF-ET)助孕中由3原核(PN)发育的废弃的优质胚胎300个,将胚胎随机分配到3个冷冻组:SSCVS组、Cryotop组和CPS组,每组为100个胚胎;通过胚胎冻融后存活率、完整胚胎存活率和囊胚形成率,比较SSCVS、Cryotop及CPS对分裂期胚胎冷冻的保存效果。结果SSCVS组和Cryotop组及CPS组的分裂期胚胎冻融后存活率、完整胚胎存活率和囊胚形成率比较,差异均有统计学意义(χ^(2)值分别为26.190、9.028、24.129,P<0.05)。SSCVS组与Cryotop组的分裂期胚胎冻融后存活率、完整胚胎存活率和囊胚形成率比较差异均无统计学意义(P>0.05);SSCVS组的分裂期胚胎冻融后存活率、完整胚胎存活率和囊胚形成率均高于CPS组(χ^(2)值分别为13.455、6.792、18.755,P<0.05);Cryotop组的分裂期胚胎冻融后存活率、完整胚胎存活率、囊胚形成率均高于CPS组(χ^(2)值分别为15.457、5.498、15.153,P<0.05)。结论通过研究建立了适合应用于临床的人分裂期胚胎的新型封闭式玻璃化冷冻系统SSCVS,其将为提高冻融胚胎妊娠率,同时预防生物污染,并提高活产率和后代安全性提供了新的技术支持。Objective To compare the cryopreservation effect of the silver-straw closed vitrification system(SSCVS),the open freezing film(Cryotop)and closed pulled straw(CPS)method on embryos at day 3 division stage,Methods A total of three hundred discarded high-quality embryos developed from 3 pronuclens(PN)in in vitro fertilization-embryo transfer(IVF-ET)assisted conception performed at the Second Hospital of West China,Sichuan University,from January 2017 to October 2018 were collected,and randomly assigned to three freezing groups:the SSCVS group,the Cryotop group and the CPS group,with loo embryos in each group.The preservation effects of SSCVS,Cryotop and CPS on the freezing of embryos at the division stage were compared by the survival rate of embryos after freezing and thawing,the survival rate of intact embryos and the rate of blastocyst formation.Results The post-freezing and thawing survival rate,intact embryo survival rate and blastocyst formation rate of split-stage embryos were compared in the SSCVS group,the Cryotop group and the CPS group,the differences were all statistically significant(χ^(2)=26.190,9.028 and 24.129,respectively,P<0.05).The differences between the SSCVS group and Cryotop group were not statistically significant(P>0.05).The survival rate after freezing and thawing,the survival rate of intact embryo,the rate of blastocyst formation in the SSCVS group were higher than those in the CPS group(χ^(2)=13.455,6.792,and 18.755,respectively,P<0.05),the survival rate after freezing and thawing,the survival rate of intactembryo,the rate of blastocyst formation in the Cryotop group were higher than those in the CPS group(χ^(2)=15.457,5.498 and 15.153,respectively,P<0.05).Conclusion A new closed vitrification freezing system SSCVS for human split-stage embryos suitable for clinical application has been established,which will provide new technical support to increase the pregnancy rate of frozen-thawed embryos,as well as to prevent biological contamination,and to improve the rate of live births and th
