摘 要:目的研究小鼠胚胎成骨前体细胞MC3T3-E1经成骨诱导后压电型机械敏感离子通道组件1(PIEZO1)的表达变化及意义。方法成骨诱导MC3T3-E1细胞后,于第0、14、21天分别进行茜素红染色观察钙结节和细胞形态,利用蛋白质印迹法(Western blot)、实时荧光定量聚合酶链反应(qRT-PCR)分别检测成骨诱导对各时间点PIEZO1蛋白和PIEZO1 mRNA表达的影响,组间比较采用One-way ANOVA检验。结果MCET3-E1成骨诱导0 d时基本未见红色钙化结节,细胞呈梭形;诱导14 d时出现钙化结节,细胞呈聚集样和层叠样生长,且随着诱导时间延长钙化结节数量增多,形状逐渐增大,细胞层叠样生长加重;同时MC3T3-E1中PIEZO1蛋白及mRNA的表达量随着成骨诱导时间的延长而逐渐增高(PIEZO1蛋白0 d组0.26±0.04、14 d组0.42±0.16、21 d组0.81±0.04,F=24.05,P<0.001;PIEZO1 mRNA 0 d组1.00±0.03、14 d组1.45±0.11、21 d组4.51±0.44,F=161.90,P<0.001),差异有统计学意义。结论成骨分化可促进MC3T3-E1在矿化结节形成期PIEZO1蛋白的表达。Objective To investigate the expression and implication of mechanosensitive ion channel Piezo type mechanosensitive ion channel component 1(PIEZO1)protein in Mouse embryo osteoblast precursor(MC3T3-E1)cells after osteogenic induction.Methods After inducing osteogenesis in MC3T3-E1 cells,calcium nodules and cell morphology were observed using alizarin red staining on day 0,14 and 21.Western blotting and real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)were used to detect the effects of osteogenic induction on PIEZO1 protein and mRNA expression at different time points.Comparison between groups was performed by One-way ANOVA test.Results On day 0 of osteogenic induction,no red calcium nodules were observed in MC3T3-E1 cells,and the cells were spindle-shaped.On day 14,calcium nodules appeared,and the cells grew in clusters and stacks.The number and size of calcium nodules increased,and the stacks of cells became more pronounced with the induction time extended.The expression of PIEZO1 protein and mRNA in MC3T3-E1 cells gradually increased with the extension of osteogenic induced time(PIEZO1 protein group 0 d 0.26±0.04,group 14 d 0.42±0.16,group 21 d 0.81±0.04,F=24.05,P<0.001;PIEZO1 mRNA group 0 d 1.00±0.03,group 14 d 1.45±0.11,group 21 d 4.51±0.44,F=161.90,P<0.001).The difference was statistically significant.Conclusion The osteogenic differentiation can promote the expression of PIEZO1 protein in MC3T3-E1 cells during the formation of mineralized nodules.
