摘  要:目的探讨和分析运动预适应对大鼠心肌一磷酸鞘氨醇(sphingosine 1-phosphate,S1P)含量的影响及其可能的机制。方法通过建立运动预适应和力竭运动动物模型,常规饲养1周后将大鼠随机分为正常对照组(C组)、运动预适应组(EP组)、运动预适应+S1P受体阻滞剂W146组(EP+W146组),运动预适应+P13K阻滞剂LY-294002组(EP+LY294002组)。C组常规饲养3周,其余各组大鼠以尾部负重3%体质量的方式进行间歇性游泳运动,运动持续3周,每周6 d,逐渐增加运动时间,第1周运动30 min/d,第3周2 h/d。3周实验后,C组、EP组、EP+W146组、EP+LY294002组大鼠进行一次性力竭游泳运动。比较各组大鼠心脏重量、心肌S1P含量及心肌细胞凋亡情况。结果与EP组相比,EP+W146组,力竭运动大鼠心脏重量增加(P<0.05)。与C组相比,EP组力竭运动大鼠S1P含量升高(P<0.05)。与C组相比,EP组、EP+W146组、EP+LY294002组力竭运动大鼠心肌凋亡指数下降(P<0.05);与EP组相比,EP+W146组、EP+LY294002组力竭运动大鼠心肌凋亡指数上升(P<0.05);与EP+W146组相比,EP组力竭运动大鼠心肌凋亡指数下降(P<0.05)。结论运动预适应可提高力竭运动大鼠心肌S1P含量,其机制可能通过P13K/Akt信号通路发挥心肌保护作用。Objective To explore and analyze the effects of exercise preconditioning on the content of myocardial sphingosine 1-phosphate(S1P)and its possible mechanisms in rats.Methods Animal models of exercise preconditioning and exhaustion were established,and the rats were randomly divided into normal control group(Group C),exercise preconditioning group(Group EP),exercise preconditioning+S1P receptor blocker W146 group(Group EP+W146),and exercise preconditioning+P13K blocker LY-294002 group(Group EP+LY294002)after 1 week of routine rearing.Group C was routinely reared for 3 weeks,and the rest of the groups were routinely reared for 3 weeks,and the rest of the groups were routinely reared for 3 weeks,and the rest of the rats were routinely reared for 3 weeks.rats were kept for 3 weeks,and the rest of rats in each group were subjected to intermittent swimming exercise with 3%of body weight on the tail,the exercise lasted for 3 weeks,6 d per week,and the exercise time was gradually increased to 30 min/d in week 1 and 2 h/d in week 3.After 3 weeks of experiments,the rats in group C,EP group,EP+W146 group,and EP+LY294002 group were subjected to a one-time exhaustion swimming exercise.The heart weight,myocardial S1P content,and cardiomyocyte apoptosis were compared among rats in each group.Results Compared with the EP group,in the EP+W146 group,the heart weight of rats undergoing forceful exercise was increased(P<0.05).Compared with group C,S1P content was increased in force-fatigue exercise rats in the EP group(P<0.05).Compared with group C,myocardial apoptosis index decreased in the EP group,EP+W146 group,and EP+LY294002 group of force-fatigue exercise rats(P<0.05);compared with the EP group,myocardial apoptosis index increased in the EP+W146 group and EP+LY294002 group of force-fatigue exercise rats(P<0.05);compared with the EP+W146 group,myocardial apoptosis index decreased(P<0.05).Conclusions Exercise preconditioning can increase myocardial S1P content in exhaustion rats,and its mechanism may play a cardioprotective role
