摘  要:目的建立南天竹根HPLC指纹图谱,分析鉴定其化学成分,并测定其总生物碱、小檗碱的含量。方法指纹图谱与小檗碱含量测定采用Agilent ODS-A色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.2%乙酸,梯度洗脱;体积流量1 mL/min;柱温30℃;检测波长365 nm。“中药色谱指纹图谱相似度评价软件”进行图谱分析。紫外分光光度法测定南天竹根总生物碱的含量。UPLC-Q-TOF-MS/MS法、Peakview软件分析鉴定化学成分。主成分分析(PCA)、判别分析(DA)对色谱峰数据进行统计学分析。结果12批南天竹根指纹图谱中有9个共有峰,相似度大于0.920。通过与数据库比对,在南天竹根总提物中鉴定出41个化合物,已知化合物32个,其中23个为生物碱。南天竹根中总生物碱、小檗碱含量差异均较大,栽培南天竹根中两者含量普遍高于野生。PCA、DA分析发现,野生、栽培南天竹成分并无差异性。结论该方法准确、快捷,可用于评价南天竹根质量。AIM To establish the HPLC fingerprints for Nandinae Radix,analyze and identify its chemical components,and determine the contents of its total alkaloids and berberine.METHODS The determination of fingerprint and berberine content were performed on a 30℃thermostatic Agilent ODS-A column(4.6 mm×250 mm,5μm)with the mobile phase comprising of acetonitrile-water(0.2%acetic acid)flowing at 1 mL/min in a gradient elution manner,and the detection wavelength was set at 365 nm.The software of similarity assessment for traditional Chinese medicines fingerprints was used for the analysis of the characteristic fingerprints.Nandinae Radix also had its total alkaloids determined by UV spectrophotometer;its chemical components analyzed and identified by UPLC-Q-TOF-MS/MS method and peak view software;and its chromatographic peaks analyzed by the principal component analysis(PCA)and discriminant analysis(DA).RESULTS A total of nine common peaks identified in the fingerprint spectra of twelve batches of Nandinae Radix shared more than 0.920 similarities.By using the the database,forty-one compounds were identified in the total extract of Nandinae Radix,with thirty-two known compounds including twenty-three alkaloids.Nandinae Radix was observed with obvious content variations in total alkaloids and berberine between the cultivated and those wild ones generally,with higher quantities found in the former.PCA and DA analyses revealed no components difference between the two different kinds of Nandinae Radix.CONCLUSION This accurate and fast method can be used for the quality assessment of Nandinae Radix.
