摘  要:目的研究醋延胡索炮制过程中原阿片碱、盐酸巴马汀、盐酸小檗碱、去氢紫堇碱、延胡索乙素、四氢小檗碱、延胡索甲素含量与色度值的相关性。方法该药物分析采用Agilent ZORBAX-Extend-C 18色谱柱(4.6 mm×250 mm,5μm);流动相0.6%乙酸-乙腈,梯度洗脱;体积流量1.0 mL/min;柱温25℃;检测波长280 nm。以不同炮制时间点醋延胡索为研究对象,比较色度值(L、a、b、E ab)与生物碱含量的相关性。结果7种生物碱在各自范围内线性关系良好(R 2≥0.9981),平均加样回收率98.35%~101.24%,RSD小于2%。炮制时间为0~15 min时,L、b、E ab与生物碱含量呈显著负相关,a与生物碱含量呈显著正相关;炮制时间为15~30 min时,L、b、E ab与生物碱含量呈显著正相关,a与生物碱含量呈显著负相关。醋延胡索在炮制过程中颜色由黄转黑时,生物碱含量升高;由黑转红时,生物碱含量降低,炮制时间为15 min时最高。结论该方法准确可行,可快速实现不同炮制时间点醋延胡索质量客观评判。AIM To study the correlation between the chromatic value and the content of protopine,palmatine chloride,berberine,dehydrocorydaline,tetrahydropalmatine,tetrahydroberberine,and corydaline during the vinegar processing of Corydalis Rhizoma.METHODS The analysis of this drug was performed on a 25℃thermostatic Agilent ZORBAX-Extend-C 18 column(4.6 mm×250 mm,5 m),with the mobile phase comprising of 0.6%acetic acid water-acetonitrile flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 280 nm.Corydalis Rhizoma being at different vinegar processing time points had its correlation between the chromatic values(L,a,b,E ab)and the contents of alkaloids assessed and compared.RESULTS Seven alkaloids showed good linear relationships within their own ranges(R 2≥0.9981),whose average recoveries were within the range of 98.35%-101.24%with less than 2%RSD.L,b,E ab found their significantly negative correlation with the alkaloid content during 0-15 minutes processing time,but significantly positive correlation during 15-30 minutes processing time;a found its significantly positive correlation with the alkaloid content in the first 15 minutes processing but negative correlation during the next 15 minutes vinegar processing.That indicated that when the color of Corydalis Rhizoma changed from yellow to black,the alkaloid content increased,and reached highest at the 15 th minute;when the color turned from black to red,the alkaloid content decreased.CONCLUSION This accurate and feasible method can quickly assess the quality of vinegar-processed Corydalis Rhizoma at different processing time points.
