摘  要:为了探讨瓠瓜(Lagenaria siceraria) MYB转录因子家族的功能,本研究基于瓠瓜的全基因组和白粉病菌侵染后的转录组数据,利用生物信息学软件对Ls MYB家族成员进行了鉴定、分析。结果表明:在瓠瓜全基因组中共鉴定到174个LsMYBs基因,其氨基酸数为103~1 673个,均为亲水性蛋白;亚细胞定位预测表明,118个LsMYB基因定位在细胞核中;根据MYB结构域重复数,可将瓠瓜MYB分为1R、2R (R2R3)、3R三个类型,其中2R类型和1R类型分别占比51.1%、47.7%;进化树分析显示,瓠瓜MYB基因家族可被分成31个亚组,推测相同亚组上的瓠瓜与拟南芥的MYB转录因子可能具有相似的功能;利用MEME软件预测得到15个motif元件,分析发现不同MYB的motif数量及分布存在一定差异,同一亚组MYB蛋白具有相似的结构特征;顺式作用元件表明,LsMYB基因启动子区域包含光响应、激素应答、生物及非生物胁迫等相关作用元件和结合位点,表明瓠瓜MYB基因家族可能广泛参与了瓠瓜的生长发育、激素调控、生物/非生物胁迫的响应;瓠瓜接种白粉病菌后MYB家族成员表达量分析表明,推测与抗白粉病相关的基因有五组:Group 1、Group 2、Group 3、Group 5、Group 6,说明这155个Ls MYB家族基因直接或间接参与了瓠瓜对白粉病的抗性。本研究为MYB基因在瓠瓜抗病育种及抗病机制的研究、应用提供了理论依据。To investigate the functions of the MYB transcription factor family in Lagenaria siceraria,this study identified and analyzed members of the LsMYB family using bioinformatics software based on the whole genome sequence and transcriptomic data after powdery mildew infestation.The results showed that a total of 174 LsMYB genes in the genome of L.siceraria,with amino acid numbers ranging from 103 to 1673,all predicted to be hydrophilic proteins.In subcellular localization predictions,118 LsMYB genes were localized in the nucleus.According to the number of MYB structural domain repeats,MYB family members in L.siceraria could be classified into 1R,2R(R2R3),and 3R types;the 2R and 1R types accounted for 51.1%and 47.7%of the MYBs,respectively.Phylogenetic analysis showed that the MYB gene family could be divided into 31 subgroups,and it was speculated that MYB transcription factors from L.siceraria and Arabidopsis in the same subgroup might have similar functions.Fifteen motif elements were predicted by MEME software,and the number and distribution of motifs were found to be different in different MYB members.Proteins in a given MYB subgroup had similar structural features.The cis-acting elements indicate that the promoter regions of LsMYB-encoding genes contain elements and binding sites related to light response,hormone response,and biotic and abiotic stresses,suggesting that the MYB gene family may be extensively involved in growth and development,hormone regulation,and biotic/abiotic stress response in L.siceraria.The expression analysis of MYB family members in L.siceraria inoculated with powdery mildew bacteria showed that there were five groups of genes associated with powdery mildew resistance:Group 1,Group 2,Group 3,Group 5,and Group 6 indicated that 155 LsMYB family genes were directly or indirectly involved in resistance to powdery mildew.This study provides a theoretical basis for research into and application of the MYB genes in breeding disease resistant L.siceraria and understanding its disease resistance
