摘 要:为明确桑天牛各发育时期的菌群结构及多样性,利用16S rRNA测序技术对桑天牛卵、初孵幼虫、老熟幼虫、雌雄成虫4个发育阶段的细菌群落进行研究。结果表明,桑天牛不同发育阶段的细菌群落多样性存在差异。初孵幼虫的Alpha多样性显著高于其他阶段。Beta多样性分析表明,雌雄成虫细菌群落高度相似。在门分类水平,卵期的优势菌门为放线菌门(64.78%),初孵幼虫期为变形菌门和厚壁菌门(38.40%、25.31%),老熟幼虫为变形菌门(93.47%),雌雄成虫为厚壁菌门(86.46%、86.35%)。在属分类水平,卵期的优势菌属为红球菌属,初孵幼虫为Muribaculaceae科未知属,老熟幼虫为Gibbsiella属,雌雄成虫为乳球菌属。在功能方面,发酵、化学异养、好氧性化学异养在桑天牛各个发育阶段的占比都较高,并且与该阶段存在的菌落有关。本研究揭示了桑天牛不同发育阶段细菌群落差异,明确了各阶段优势肠菌,可为桑天牛的生物防治奠定基础。In order to clarify the microflora structure and diversity of Apriona germari at different developmental stages,the bacterial communities at four developmental stages of A.germari were studied,including eggs,newly hatched larvae,mature larvae,male adults,and female adults.The 16S rRNA sequencing technology was used to analyze the bacterial communities of A.germari.The results showed that there were differences in bacterial community diversity at different developmental stages.The newly hatched larvae showed higher diversity than that at other three developmental stages.Beta diversity analysis showed that the bacterial communities of male and female adults were similar.In terms of phylum classification level,Actinobacteriota was the dominant bacteria in the egg stage(64.78%),Proteobacteria and Firmicutes in the newly hatched larvae(38.40%,25.31%),Proteobacteria in the mature larvae(93.47%),Firmicutes in male and female adults(86.46%,86.35%).In terms of genus classification level,Rhodococcus was the dominant bacterial genus in egg stage,Muribaculaceae and unknown genus in the newly hatched larvae,Gibbsiella in the mature larvae,and Lactococcus in the male and female adults.In terms of function,such as fermentation,chemical heterotrophy and aerobic chemical heterotrophy were mainly high proportions in different stages,and related to the existence of colonies in each stage.This study revealed the differences of bacterial communities in different developmental stages of A.germari and identified the dominant intestinal bacteria at each stage,which could lay the foundation for biological control of A.germari.
