摘 要:阐明罗伊氏乳杆菌FLRE5K1可以改善对乙酰氨基酚(APAP)诱导的高胆红素血症。对昆明鼠连续7 d灌胃L.reuteri FLRE5K1,7 d后,实验组小鼠腹腔注射500 mg·Kg^(-1)的APAP造模,对小鼠禁食不禁水12 h,之后眼球取血用于测定血清生化指标,并对其肝脏进行组织病理学分析。结果表明L.reuteri FLRE5K1能够降低小鼠血清中总胆红素、尿素以及肌酐含量。病理切片分析也表明,L.reuteri FLRE5K1对APAP引起的炎症有明显的缓解作用。通过分子检测,发现L.reuteri FLRE5K1能提高肝脏中多药耐药相关蛋白2(MRP2)表达水平而影响胆红素代谢,改善高胆红素血症,填补了目前益生菌在改善高胆红素血症机制探究方面的空白,为未来临床辅助治疗提供技术支撑,具有较好的潜在应用价值。It was suggested that L.reuteri FLRE5K1 can ameliorate acetaminophen(APAP)-induced hyperbilirubinemia.The Kunming mice were administrated intragastrically with L.reuteri FLRE5K1 for 7 consecutive days.After 7 days,the experimental mice were intraperitoneally injected with 500 mg/Kg APAP to establish the model.Then the mice were fasted but allowed access to water for 12 h.The results showed that L.reuteri FLRE5K1 could reduce the contents of total bilirubin,urea and creatinine in serum of mice.The analysis of pathological sections also showed that L.reuteri FLRE5K1 had a significant alleviating effect on the inflammation caused by APAP.Through molecular detection,it was found that L.reuteri FLRE5K1 could improve the expression level of Multidrug resistance associated protein 2(MRP2)in the liver,thus affecting the metabolism of bilirubin and relieving hyperbilirubinemia.This study fills the gap in the exploration of the mechanism of improving hyperbilirubinemia by probiotics,provides technical support for future adjuvant clinical therapy,and has good potential application value.
