摘  要:目的 分析五味子及不同炮制品炮制后化学成分差异,为五味子不同炮制品的整体质量控制提供依据。方法 建立生五味子、醋五味子、酒五味子指纹图谱,进行相似度评价,同时进行主成分分析(PCA分析)及正交偏最小二乘法判别分析(OPLS-DA分析),结合网络药理学分析探讨差异成分对应的靶点和通路。结果 建立的指纹图谱中成功指认出21个共有峰,其中4个峰分别被指认为五味子醇甲、五味子酯甲、五味子甲素和五味子乙素;相似度结果显示,五味子不同炮制品指纹图谱与各自对照图谱的相似度在0.907~0.999。PCA和OPLS-DA结果表明,五味子不同炮制方法对五味子活性成分的含量变化有一定的影响。网络药理学分析表明,指认出的色谱峰与五味子的主要药理作用密切相关。结论 所建立的指纹图谱方法精密度、重复性、稳定性良好,结合多元统计方法和网络药理学分析,为五味子及其炮制品质量标准评价提供了科学依据。Objective To analyze differences in chemical composition of Wuweizi(Schisandrae Chinensis Fructus)and different processed products after preparation,which provide a basis for the overall quality control of different Wuweizi(Schisandrae Chinensis Fructus)processed products.Methods Wuweizi(Schisandrae Chinensis Fructus)fingerprints of raw,vinegar and wine were established,similarity evaluation was carried out,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were carried out at the same time,and the corresponding targets and pathways of different components were explored in combination with network pharmacological analysis.Results In the established fingerprint,21 common peaks were successfully identified,of which 4 peaks were identified as schizandrol A,schisantherin A,schizandrin A,schizandrin B;The similarity results showed that the similarity between the fingerprint of different Wuweizi(Schisandrae Chinensis Fructus)processed products and their respective control maps was 0.907-0.999.The results of PCA and OPLS-DA showed that different preparation methods of Wuweizi(Schisandrae Chinensis Fructus)had a certain influence on the content of Wuweizi(Schisandrae Chinensis Fructus)active ingredients.Network pharmacological analysis showed that the identified chromatographic peaks were closely related to the main pharmacological effects of Wuweizi(Schisandrae Chinensis Fructus).Conclusion The established fingerprint method has good precision,repeatability and stability,combined with multivariate statistical method and network pharmacological analysis,which provides a scientific basis for the evaluation of quality standards of Wuweizi(Schisandrae Chinensis Fructus)and its artillery products.
