摘　　要：目的探讨不同生长年限的白及、小白及和黄花白及药材中1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯(militarine,化学式为C_(34)H_(46)O_(17))成分的积累规律,为白及质量控制和近缘种资源的开发提供依据。方法采用HPLC法,以XTerra Waters柱(5μm,4.6 mm×250 mm)为色谱柱,以乙腈-0.1%磷酸溶液(22∶78)进行梯度洗脱,流速为1.0 mL/min,检测波长270 nm,测定各样品militarine的含量。结果经过对HPLC图谱分析比较,不同生长年限的白及与其2种近缘种的militaeine成分含量差别较大,以4年生最高,3年生的次之,2年生则最低,相同生长年限的种间含量稍有差异,白及稍高,小白及与黄花白及稍低。结论以3年及以上生长年限的白及作为药材原料具有一定的科学性。小白及、黄花白及与生长年限相同的白及的militarine含量相当,二者作为习用品具有一定的合理性,可为开展深入的资源评价提供参考依据。Objective To explore the accumulation regularity of militarine components in Baiji(Bletilla striata),Xiaobaiji(Bletilla formosana)and Huanghua Baiji(Bletilla ochracea)with different growth years,in order to provide basis for quality con-trol of Baiji(Bletilla striata)and development of related species resources.Methods The chromatographic separation was per-formed on XTerra Waters(5μm,4.6 mm×250 mm)column.The mobile phase consisting of acetonitrile-phosphoric acid(0.1%)solution(22∶78)in gradient elution.The flow rate was 1.0 mL·min^(-1),and the detection wavelength was 270 nm.Re-sults Through HPLC chromatographic analysis,the contents of militaeine in Baiji(Bletilla striata)and its two related species with different growth years were quite different,with the highest content in 4-year-old,the second in 3-year-old and the lowest in 2-year-old Baiji(Bletilla striata).The contents of militaeine in Baiji(Bletilla striata),Xiaobaiji(Bletilla formosana)and Huanghua Baiji(Bletilla ochracea)with the same growth years were slightly different.The contents in Baiji(Bletilla striata)were slightly higher,those in Xiaobaiji(Bletilla formosana)and Huanghua Baiji(Bletilla ochracea)were slightly lower.Conclusion It is scientific to use Baiji(Bletilla striata)which is 3 years old or above as the raw material.Baiji(Bletilla striata),Xiaobaiji(Bletilla formosana)and Huanghua Baiji(Bletilla ochracea)have little difference in the content of militarine with the same growth years.The latter two are reasonable as folk products,which can provide a reference for in-depth resource evaluation.
