摘 要:目的研究旨在探讨痤疮合剂对辅助性T细胞(Helper T Cell,Th)1、Th2、Th17、调节性T细胞(Regulatory T cell,Treg)平衡的调节作用。方法除对照组,其他干预组用Kligman造模法制造兔耳痤疮模型2周,取病理,判断造模成功后,痤疮合剂组灌服痤疮合剂17.52 g/(kg·d);阳性药物组灌服清热暗疮胶囊0.12 g/(kg·d);对照组及模型组灌服等量生理盐水,1次/d,共30 d。取静脉血及患处组织、耳根淋巴结,苏木精-伊红染色法(Hematoxylin-eosin staining,HE)检测毛囊面积和皮脂腺直径。酶联免疫吸附测定法(Enzyme linked immunosorbent assay,ELISA)检测血清中干扰素(Interferon,IFN)-γ、白介素(Interleukin,IL)-5、IL-17、IL-12、IL-22、IL-10、转化生长因子(Transforming growth factor,TGF)-β含量。蛋白质印迹法(Western blot)、免疫组织化学法检测耳根淋巴结中T盒转录因子(T-box expressed in T cell,T-bet)、GATA结合蛋白3(GATA binding protein 3,GATA3)、视黄酸受体相关孤儿核受体γt(Retineic-acid-receptor-related orphan nuclear receptor gamma,RORγt)、叉头盒转录因子p3(Forkhead box P3,Foxp3)表达水平。结果痤疮合剂能明显改善兔耳表皮层的结节、脓疱、组织厚度、囊肿及毛囊口扩张程度,减少炎症细胞的增殖和浸润,表皮和内壁角质层变薄松软。与对照组相比,其余各组IFN-γ、IL-12、IL-17、IL-22含量显著增高,IL-5、TGF-β、IL-10含量显著降低(P<0.01)。与模型组比较,痤疮合剂和阳性组IFN-γ、IL-17、IL-12、IL-22含量明显下降,IL-5、TGF-β、IL-10含量显著增高(P<0.01)。与对照组比较,其余各组T-bet、RORγt的表达及平均光密度值(Integral-Optimistically,IOD)显著增大,GATA3、Foxp3表达及IOD下降(P<0.01)。与模型组相比,痤疮合剂和阳性组T-bet、RORγt表达及IOD下调,GATA3、Foxp3表达及IOD上调(P<0.01)。结论痤疮合剂通过调控炎性因子平衡Th1/Th2、Th17/Treg的极化偏移,实现免疫细胞内稳态,改善皮损病理表现,对�Objective To study the regulating effect of Acne Mixture(痤疮合剂)on the balance of helper T cell(Th)1/Th2 and Th17/regulatory T cell(Treg).Methods Except the control group,the other three intervention groups used Kligman method to prepare the rabbit ear acne model for 2 weeks.After successfully established the animal model,the Acne Mixture group was given Acne Mixture[17.52 g/(kg·d)].The positive drug group was given Qingre Anchuang Capsule(清热暗疮胶囊)[0.12 g/(kg·d)].The control group and the model group were given the equal amount of saline once a day,continuous dosing for 30 d.The venous blood,the affected tissues and the ear root lymphoid nodes were collected.Hematoxylin-eosin staining(HE)staining was used to detect the hair follicle and the sebaceous gland diameter.ELISA was used to determine the serum contents of interferon(IFN)-γ、interleukin(IL)-5、IL-17、IL-12、IL-22、IL-10 and transforming growth factor(TGF)-β.Western blot and immunohistochemistry were used to detect the expression levels of T-box expressed in T cell(T-bet),GATA binding protein 3(GATA3),etineic-acid-receptor-related orphan nuclear receptor gamma(RORγt)and forkhead box P3(Foxp3)in the ear root lymph nodes.Results Acne Mixture improved the nodules,pustules,tissue thickness,cysts and hair follicle expansion degree of rabbit ear epidermis.It also reduced the proliferation and infiltration of inflammatory cells,and the epidermis and inner cuticle became thinner and softer.Compared with those of the control group,the contents of IFN-γ,IL-17,IL-12 and IL-22 in other three groups were significantly increased,while the contents of IL-5,TGF-βand IL-10 were significantly decreased(P<0.01).Compared with those of the model group,the contents of IFN-γ,IL-17,IL-12 and IL-22 were decreased in the Acne Mixture group and the positive drug group,while the contents of IL-5,TGF-βand IL-10 were increased(P<0.01).Compared with those of the control group,the expressions of T-bet and RORγt and the mean integral optical density(IOD)in
