摘 要:目的 研究不同死亡时间(PMI)大鼠肝脏浸渍液电导率(EC)及其相关化学小分子的含量与晚期PMI的关系,从而探讨应用组织EC推断PMI的可能性以及EC的内在变化机制。方法 选用48只成年健康无病原体SD大鼠,在同一时间处死后保存于25℃左右的恒温环境中,分别于死后7 d内提取大鼠肝组织并制成匀浆浸渍液,测定浸渍液EC及其常见小分子(电解质、微量元素、无机氮)的含量,并分析肝组织EC、导电小分子以及PMI三者之间的关系。结果 大鼠死后7 d内,随着PMI的延长,肝脏的EC明显增加,钙离子、镁离子、无机磷、尿素氮和肌酐的含量也随着PMI的增加而明显增加R^(2)=(0.93~0.99)。钾离子和尿酸的含量随着PMI的延长而上下波动,其变化没有统计学意义(P> 0.05)。死亡1 d后,没有检测到钠离子和氯离子。此外,钙离子、镁离子、无机磷、尿素氮和肌酐与EC的变化具有显著的线性关系,R^(2)=(0.87~0.98)。结论 利用EC和多种肝脏化学小分子的含量变化来推断晚期PMI有潜在的应用价值。而且,这些化学小分子含量的变化也可能有助于理解死后肝脏分解过程中EC增加的微观机制。Objective To investigate the relationship of electrical conductivity(EC)and its related small chemical molecules in rat liver with late postmortem interval(PMI),so as to explore the possibility of applying tissue EC to infer PMI and the underlying mechanism.Methods Forty-eight adult healthy pathogen-free SD rats were executed at the same time and stored at a constant temperature of about 25°C.The liver tissues of rats were extracted and homogenized into macerates within 7 d after death.The EC of the liver macerate and the levels of small chemical molecules(electrolytes,trace elements,inorganic nitrogen)were determined to analyze the association of EC and small chemical molecules with PMI.Results Within 7 days after death,the EC of the liver macerate and the contents of calcium,magnesium,inorganic phosphorus,urea nitrogen and creatinine increased significantly with the increase of PMI R?=(0.93~0.99).The levels of potassium and uric acid fluctuated up and down with the increase of PMI and the changes showed no statistical significance(P>0.05).Sodium and chloride levels of liver was not detected within 1 d after death.In addition,calcium,magnesium,inorganic phosphorus,urea nitrogen and creatinine had a significant liner relationship with EC,R’=(0.87~0.98).Conclusion It has potential application value to infer late PMI by using the content changes of EC and variety small chemical molecules in liver.Furthermore,changes in the levels of these small chemical molecules may also help to understand the microscopic mechanisms of EC increase during postmortem liver decomposition.
