基于流式细胞法和基因组K-mer分析法检测漆树基因组大小

摘　　要：漆树Toxicodendron vernicifluum(Stokes)F.A.Barkl.是我国重要的经济林木资源,在工业、药用、生态、文化等领域产生了积极的影响。本研究利用流式细胞技术和高通量测序技术对漆树基因组特征进行了初步研究。利用已知基因组大小的柳树(Salix matsudana Koidz)作为对照,利用流式细胞技术估算漆树基因组大小为538.90 Mb。利用高通量测序技术对漆树基因组进行K-mer分析,估算漆树基因组大小为605.9Mb,基因组杂合度为0.572%,重复序列约占71.4%,GC含量为37.2%。从基因组基本结构特征上看,漆树基因组属于高重复、高杂合的复杂基因组。利用获得的DNA序列数据,从头组装了一个大小为638.5Mb,contig N50长度为153bp的漆树基因组。本研究为漆树全基因组精细图谱的绘制和分子育种研究提供了重要的理论依据。Lacquer tree(Toxicodendron vernicifluum(Stokes)F.A.Barkl.)is an important economic forestry resource in China and play an important role in industrial,medicinal,ecological,and cultural fields.In this study,we conducted preliminary research on the genomic characteristics of lacquer tree using both flow cytometry and high-throughput sequencing techniques.Using willow(Salix matsudana Koidz)with known genome size as a control,the lacquer genome size was estimated to be 538.90Mb by flow cytometry.K-mer analysis of the lacquer genome by using high-throughput sequencing technology estimated that the genome size to be 605.9Mb,genome heterozygosity to be 0.572%,repetitive sequences to be about 71.4%,and the GC content to be 37.2%.From the basic structural characteristics of the genome,the genome of lacquer tree belongs to a complex genome with high duplication and heterozygosity.Using the obtained DNA sequence data,we assembled a de novo genome.The genome is 638.5Mb,with an N50 size of 153 bp.This study provides an important theoretical basis for the construction of a fine-scale map of the lacquer tree and molecular breeding research.
