摘  要:目的:探究胰高血糖素样肽-1(GLP-1)类似物司美格鲁肽对链脲佐菌素(STZ)诱导的糖尿病大鼠认知功能的影响。方法:实验分为正常组(NC组)、糖尿病组(DM组)、司美格鲁肽治疗组(DM+SE组),构建糖尿病大鼠模型,Morris水迷宫实验评价大鼠空间学习与记忆相关认知功能,腹主动脉采血检测大鼠血糖、血脂、血Betatrophin与血胰岛素水平。结果:DM组空腹血糖水平高于NC组(P=0.001),DM+SE组甘油三酯水平高于NC组(P=0.006);水迷宫定位航行实验,第3天DM组(P=0.010)、第4天DM组(P=0.006)与DM+SE组(P=0.011)、第5天DM组(P=0.002)与DM+SE组(P=0.014)逃避潜伏期均长于NC组;DM组(P<0.001)与DM+SE组(P<0.001)Betatrophin水平较NC组明显升高。结论:司美格鲁肽不能改善糖尿病大鼠空间学习与记忆相关认知功能障碍,可能与其使用剂量相关,Betatrophin可调节糖代谢促进糖尿病的发生,但不影响胰岛素分泌。Objective:To investigate the effect of semaglutide,an analog of glucagon like peptide-1(GLP-1),on cognitive function in streptozotocin(STZ)induced diabetic rats.Methods:The experiment includes normal group(NC group),diabetes group(DM group)and semaglutide treatment group(DM+Se group).The diabetic rat model was constructed.Morris water maze test was used to evaluate the spatial learning and memory cognitive function of rats.Blood glucose,blood lipid,blood betatrophin and blood insulin levels were detected by abdominal aorta blood sampling.Results:The fasting blood glucose level in DM group was higher than that in NC group(P=0.001),and the triglyceride level in DM+Se group was higher than that in NC group(P=0.006);In the Morris water maze test,the escape latency of DM group(P=0.010)on the third day,of DM group(P=0.006)and DM+Se group(P=0.011)on the fourth day,of DM group(P=0.002)and DM+Se group(P=0.014)on the fifth days was longer than that of NC group;Betatrophin levels in DM group(P<0.001)and DM+Se group(P<0.001)were significantly higher than those in NC group.Conclusion:Semaglutide can not improve the cognitive impairment related to spatial learning and memory in diabetic rats,which may be related to the dosage.Betatrophin can regulate glucose metabolism and promote the occurrence of diabetes,but does not affect insulin secretion.
