摘  要:目的:通过麻荆止咳颗粒调控胸腺基质淋巴细胞生成素(Thymic Stromal Lymphopoietin,TSLP)/蛋白酶激活受体2(Proteinase Activated Receptor-2,PAR2)/瞬时电位感受器香草酸受体1(Transient Receptor Potential Vanilloid 1,TRPV1)/活化T细胞核因子(Nuclear Factor of Activated T cells,NFAT)信号通路的动物实验,探讨其治疗咳嗽变异性哮喘(Cough Variant Asthma,CVA)的疗效机制。方法:将豚鼠随机分为空白对照组、模型组、中药组和地塞米松组,构建CVA豚鼠模型,药物干预10天,采用辣椒素激发各组豚鼠,观察咳嗽情况;苏木精-伊红(Hematoxylin-Eosin,HE)染色法观察各组肺组织病理学变化;酶联免疫吸附试验(Enzyme-Linked Immunosorbent Method,ELISA)法检测各组血清TSLP含量;免疫组化染色及实时定量聚合酶链反应(quantitative Real-time Polymerase Chain Reaction,qRT-PCR)法检测肺组织PAR2、TRPV1、NFAT的表达。结果:与模型组对比,中药组和地塞米松组药物干预后咳嗽次数明显减少;血清TSLP含量显著降低;且肺组织病理变化较模型组明显减轻;PAR2、TRPV1和NFAT的蛋白与mRNA表达均下调。结论:麻荆止咳颗粒可通过抑制CVA模型豚鼠TSLP/PAR2/TRPV1/NFAT信号通路表达,降低气道敏感性及改善气道炎症,达到止咳效果。Objective:To investigate the efficacy mechanism of the Majing Zhike granules(麻荆止咳颗粒)on cough variant asthma(CVA)by modulating thymic stromal lymphopoietin(TSLP)/proteinase activated receptor-2(PAR2)/transient receptor potential vanilloid 1(TRPV1)/nuclear factor of activated T cells(NFAT)signaling pathway through the animal experiment.Methods:Guinea pigs were randomly divided into the blank control group,the model group,the TCM medicine group andO the dexamethasone group.The guinea pig model of CVA was established.After 10 days of drug intervention,the guinea pigs in each group were stimulated with capsaicin to observe the cough.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of lung tissue in each group.Serum TSLP was detected by enzyme-linked immunosorbent method(ELISA),immunohistochemical staining and quantitative real-time polymerase chain reaction(qRT-PCR)were used to detect the expression of PAR2,TRPV1 and NFAT.Results:Compared with the model group,the number of coughs was significantly reduced in the TCM medicine group and the dexamethasone group after drug intervention,serum TSLP levels were significantly lower,and lung histopathological changes were significantly reduced,and protein and mRNA expressions of PAR2,TRPV1,NFAT were down-regulated.Conclusion:The Majing Zhike granules can inhibit TSLP/PAR2/TRPV1/NFAT signal pathway in CVA guinea pig model,reduce the sensitivity and inflammation of airway sensitivity,and achieve the effect on relieving cough.
