摘 要:目的:基于网络药理学探讨贯叶金丝桃治疗慢性前列腺炎的作用机制。方法:通过查询中药系统药理学数据库与分析平台(TCMSP)与本草组鉴网站(HERB)分别获取贯叶金丝桃的活性成分及作用靶点,通过人类基因数据库(GeneCards)、在线人类孟德尔遗传数据库(OMIM)、DisGenet数据库获取慢性前列腺炎的作用靶点,运用Venny 2.1.0平台得到贯叶金丝桃与慢性前列腺炎的交集靶点,运用Cytoscape软件构建成分-靶点网络,筛选核心成分;以STRING平台数据为基础,运用Cytoscape软件对交集靶点构建蛋白质-蛋白质相互作用网络,筛选核心靶点;运用David平台对交集靶点进行基因本体论(GO)及京都基因与基因组百科全书(KEGG)富集分析。结果:获得贯叶金丝桃活性成分6个、作用靶点193个,慢性前列腺炎的相关靶点8940个,映射后得到贯叶金丝桃作用于慢性前列腺炎的潜在靶点185个,相关度较高的靶点为肿瘤蛋白p53(Tumor Protein p53,TP53)、JUN原癌基因(Jun Proto-Oncogene,JUN)、丝裂原活化蛋白激酶(Mitogen-Activated Protein Kinase,MAPK)3、丝氨酸/苏氨酸蛋白激酶1(Akt Serine/Threonine Kinase 1,AKT1)、雌激素受体1(Estrogen receptor 1,ESR1)、MAPK1、肿瘤坏死因子(Tumor Necrosis Factor,TNF),主要富集在调控糖尿病并发症的晚期糖基化终末产物(Advanced Glycation End Products,AGE)-AGE受体(AGE Receptor,RAGE)信号通路、白细胞介素(Interleukin,IL)-17信号通路、TNF信号通路。结论:贯叶金丝桃可能通过抑制炎症反应、提高抗氧化能力、抑制抑郁过程减少雌激素分泌,从而治疗慢性前列腺炎。Objective:To explore the action mechanism of Guanyejinsitao(Herba Hyperici Perforati)on chronic prostatitis based on network pharmacology.Methods:The active components and action targets of Guanyejinsitao were obtained from TCMSP database and HERB website,and the action targets of chronic prostatitis were obtained by GeneCards,OMIM and DisGenetO databases.The intersection target of Guanyejinsitao and chronic prostatitis was obtained by Venny 2.1.0 platform.The component-target network was constructed and core components were screened by Cytoscape software.Based on STRING platform data,the protein-protein interaction network was constructed by Cytoscape software to screen the core targets.David platform was used for gene ontology analysis and Kyoto Encyclopedia of Genes and Genomes enrichment analysis of intersection targets.Results:There were 6 active components and 193 action targets of Guanyejinsitao.There were 8940 targets related to chronic prostatitis,and 185 potential targets of Guanyejinsitao actiong on chronic prostatitis after mapping.The targets with high correlation were TP53,JUN,MAPK3,AKT1,ESR1,MAPK1 and TNF,which were mainly enriched in AGE-RAGE signaling pathway in diabetic complications,IL-17 signaling pathway,TNF signaling pathway and so on.Conclusion:Guanyejinsitao may treat chronic prostatitis by inhibiting inflammatory reaction,enhancing antioxidant capacity,inhibiting the process of depression,and reducing estrogen secretion.
