摘  要:目的:建立一种简便易行的中药活性成分筛选方法,为质量标志物的确定提供可靠的实验基础。方法:以中空纤维为载体,使其壁孔填充庚醇、内腔充满磷脂酰胆碱特异性磷脂酶C(phosphatidylcholine-specific phospholipase,PC-PLC)溶液,构建中空纤维靶标捕获平台,从姜黄药材提取液中筛选潜在的PC-PLC抑制剂,结合高效液相色谱法对姜黄质量标志物进行初步辨识。结果:从不同产地的姜黄药材中均捕获到3种活性化合物,分别为双去甲氧基姜黄素、去甲氧基姜黄素和姜黄素。结论:双去甲氧基姜黄素、去甲氧基姜黄素和姜黄素可作为潜在的姜黄质量标志物;该研究建立的中空纤维靶标捕获法成本低、操作简便、捕获效率高,为中药质量标志物辨识提供了一种有效的方法。OBJECTIVE To ascertain and verify quality marker of turmeric(Curcumae Longae Rhizoma)by developing a convenient screening method of active ingredients.METHODS The target of phosphatidylcholine specific phospholipase C(PC-PLC)solution was loaded in the lumen of hollow fiber and heptanol was impregnated in the wall pores of hollow fiber,which was applied to preliminarily screen and identify potential enzyme inhibitor from turmeric extract combined with high performance liquid chromatography.RESULTS Three active compounds(including bisdemethoxy curcumin,demethoxycurcumin and curcumin)were obtained in turmeric from different producing areas.CONCLUSION The three curcuminoids can be used as potential quality markers of turmeric.The established hollow fiber target fishing method showed low cost,convenient operation and high screening ability,which provided an efficient method for the identification of quality marker in traditional Chinese medicines.
