摘 要:目的:研究小红参对高胆碱饮食C57BL/6J小鼠肠屏障功能及炎症反应的影响。方法:将18只C57BL/6J小鼠随机分为空白对照组、高胆碱饮食组和小红参治疗组,连续给药6周后,苏木精-伊红染色法(HE)镜检观察结肠绒毛长度及结肠病理变化;酶联免疫检测法检测血清中D-乳酸、二胺氧化酶(diamine oxidase,DAO)、髓过氧化物酶(myeloperoxidase,MPO)含量;Western blot法检测肠组织中紧密连接蛋白ZO-1、Occludi、Claudin-1的表达;RT-PCR法检测结肠组织中炎症相关因子肿瘤坏死因子α(tumor necrosis factor-alpha,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)的表达水平。结果:小红参治疗后能增加小鼠肠绒毛长度,降低血清因子D-乳酸、DAO、MPO水平,上调紧密连接蛋白ZO-1、Occludin、Claudin-1的表达,抑制炎症反应及炎症因子TNF-α、IL-1β、IL-6的表达。结论:小红参可能通过修复结肠屏障功能和减轻炎症反应,达到改善高胆碱饮食小鼠肠道屏障损伤的作用。OBJECTIVE Tostudy the effects of Rubia yunnanensis(Franch.)on intestinal barrier function and inflammatory response in high choline diet C57BL/6J mice.METHODS Eighteen C57BL/6J mice were randomly divided into blank control group,high choline diet group and R.yunnanensis(Franch.)treatment group.After 6 weeks of continuous administration,the colonic villus length and pathological changes of colon were observed by hematoxylin-eosin staining(HE).The contents of D-lactic acid,diamine oxidase(DAO)and myeloperoxidase(MPO)in serum were detected by Enzyme-linked immunosorbent assay(ELISA).Tight junction proteins zona occulden-1(ZO-1),Occludin and Claudin-1 in colon tissue were detected by Western blot.The expression levels of inflammation-related factors tumor necrosis factor-alpha(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)in colon tissues were detected by RT-PCR.RESULTS After treatment with R.yunnanensis(Franch.),the length of intestinal villus was increased,the levels of serum factors D-lactic acid,DAO and MPO were decreased,the expression of tight junction proteins ZO-1,Occludin and Claudin-1 was up-regulated,and the expression of inflammatory factors TNF-α,IL-1βand IL-6 were inhibited.CONCLUSION R.yunnanensis(Franch.)may ameliorate intestinal barrier damage in mice with high choline diet by repairing colon barrier function and reducing inflammatory response.
