摘　　要：目的 为了延长碱蓬多糖在体内滞留时间,选取薄膜分散-挤出法制备碱蓬多糖长循环脂质体。方法 以碱蓬多糖长循环脂质体制备过程中的磷脂浓度(mg/mL)、PEG_(2000)-DSPE浓度(mg/mL)、磷脂与药物的质量比和磷脂与胆固醇的质量比为考察对象,以包封率(%)为评价指标,采用Box-Behnken效应面法对碱蓬多糖长循环脂质体的处方进行优化。结果 碱蓬多糖长循环脂质体的包封率为88.25%,体外释放试验结果显示,与碱蓬多糖相比,碱蓬多糖长循环脂质体有一定的缓释作用。结论 采用响应面法对碱蓬多糖长循环脂质体的制备条件进行优化,并对碱蓬多糖长循环脂质体进行体外释放考察,证明该方法具有良好的可行性。Objective In order to prolong the retention time of Suaeda salsa polysaccharide in vivo,the film dispersion-extrusion method was used to prepare Suaeda salsa polysaccharide long-circulating liposomes.Methods The concentration of phospholipid(mg/mL),concentration of PEG2000-DSPE(mg/mL),mass ratio of phospholipid to drug and mass ratio of phospholipid to cholesterol were investigated during the preparation of Suaeda salsa polysaccharide long-circulating liposomes,and the encapsulation rate(%)was used as the evaluation index.The Box-Behnken response surface method was used to optimize the formulation of long circulating liposomes of the polysaccharide.Results The encapsulation rate of Suaeda salsa polysaccharide long-circulating liposomes was 88.25%.The results of in vitro release test showed that Suaeda salsa polysaccharide long-circulating liposomes had a certain slow release effect compared with Suaeda salsa polysaccharide.Conclusion The response surface methodology was used to optimize the preparation conditions of Suaeda salsa polysaccharide long-circulating liposomes,and the in vitro release of Suaeda salsa polysaccharide long-circulating liposomes was investigated,demonstrating the good feasibility of this method.
