摘　　要：目的:观察二氢槲皮素(DHQ)对心力衰竭(HF)大鼠心功能的影响,并分析其相关作用机制。方法:将45只以腹腔注射盐酸阿霉素法造模的HF大鼠随机分为模型组(HF组)、DHQ低剂量组(L-DHQ组)、DHQ高剂量组(H-DHQ组)、卡托普利组(Captopril组),每组15只。另取15只健康大鼠设为对照组(NC组)。以彩色超声仪检测大鼠心功能指标;苏木精-伊红(HE)染色观察大鼠心肌组织病理变化;脱氧核糖核苷酸末端转移酶介导的缺口末端标记(TUNEL)染色法检测大鼠心肌细胞凋亡情况;酶联免疫吸附试验(ELISA)法检测大鼠血清中脑钠肽(BNP)、炎性因子、氧化应激指标水平;蛋白免疫印迹(Western Blot)法检测大鼠心肌组织相关蛋白表达。结果:与NC组比较,HF组大鼠心肌组织存在明显的病理损伤,心功能指标左室射血分数(LVEF)、左室短轴缩短率(LVFS)、每搏心输出量(SV)均降低,BNP水平升高,血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)、丙二醛(MDA)水平均升高,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平均降低,心肌细胞凋亡率、心肌组织中B淋巴细胞瘤-2相关蛋白(Bax)、切割型半胱氨酸天冬氨酸蛋白水解酶-3(Cleaved Caspase-3)、高迁移率族蛋白1(HMGB1)蛋白表达量以及磷酸化酪氨酸激酶2(p-JAK2)/酪氨酸激酶2(JAK2)和磷酸化信号转导与转录激活因子3(p-STAT3)/信号转导与转录激活因子3(STAT3)比值均升高,B淋巴细胞瘤-2(Bcl-2)蛋白表达量降低,差异均有统计学意义(P<0.05)。与HF组比较,L-DHQ组、H-DHQ组和Captopril组大鼠心肌组织病理损伤明显减轻,LVEF、LVFS、SV均升高,BNP水平降低,血清IL-6、TNF-α、CRP、MDA水平均降低,SOD、GSH-Px水平均升高,心肌细胞凋亡率、心肌组织中Bax、Cleaved Caspase-3、HMGB1蛋白表达量以及p-JAK2/JAK2和p-STAT3/STAT3比值均降低,Bcl-2蛋白表达量升高,且H-DHQ组和Captopril组改善程度均明显优于L-DHQ组,差�Objective:To investigate the effect of dihydroquercetin(DHQ)on cardiac function in rats with heart failure(HF)and analyze the related mechanism.Methods:Forty-five HF rats were randomly divided into model group(HF group),low-dose DHQ group(L-DHQ group),high-dose DHQ group(H-DHQ group),and captopril group,with 15 rats in each group.Another 15 healthy rats were set as control group(NC group).The indexes of cardiac function were measured by color ultrasound.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of rat myocardium.Terminal deoxynucleotidyl transferase-mediated nick end labeling(TUNEL)staining was used to detect the apoptosis of rat cardiomyocytes.The levels of brain natriuretic peptide(BNP),inflammatory factors,and oxidative stress were detected by enzyme-linked immunosorbent assay(ELISA).The expression of related proteins in rat myocardium was detected by Western Blot.Results:Compared with NC group,the myocardial tissue of rats in HF group showed obvious pathological injury,left ventricular ejection fraction(LVEF),left ventricular fraction shortening(LVFS),and stroke volume(SV)decreased,and B-type natriuretic peptide(BNP)level increased;the levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),C-reactive protein(CRP),and malonaldehyde(MDA)in rat serum increased,while the levels of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in rat serum decreased;the apoptosis rate of cardiomyocytes,the expression of Bax,cleaved Caspase-3,high mobility group box 1(HMGB1)protein and the ratio of phosphorylation Janus kinase 2(p-JAK2)/JAK2 and phosphorylation signal transducer and activator of transcription 3(p-STAT3)/STAT3 increased,while the expression of Bcl-2 protein decreased;the differences waere statistically significant(P<0.05).Compared with HF group,the pathological injury of myocardial tissue in L-DHQ group,H-DHQ group,and captopril group significantly reduced;LVEF,LVFS,and SV increased,BNP level was decreased;the levels of IL-6,TNF-α,CRP,and MDA in rat serum decrea
