摘  要:本文采用荧光标记扩增片段长度多态性技术(fluorescent amplified fragment length polymorphism,fAFLP)对燕山地区出口特产果品京东板栗的10个群体269个样本进行遗传多样性分析。结果显示,共扩增位点383个,位点大小在70~500 bp范围内,其中多态性位点为314个,多态性比例为81.89%;10个群体观测等位基因数、有效等位基因数、Nei’s多样性指数、Shannon信息指数平均值分别为1.4573±0.0905、1.3101±0.0611、0.1702±0.0297和0.2514±0.0487;10个地理群体遗传距离在0.0454~0.1563之间,相似系数在0.8234~0.9786之间,UPGMA聚类得出京东板栗亲缘关系图谱。研究我国主要出口京东板栗群体的遗传结构,以期为合理评估和保护其种质资源、开展遗传育种以及提升产品品质维护板栗国际市场声誉提供基础资料。In this paper,fluorescent amplified fragment length polymorphism(fAFLP)labeling technology was used to amplify and analyze the genetic structure of 269 samples from 10 populations of Jingdong chestnuts a specialized nut export from Yanshan area.The results showed that there were 383 amplified loci in the range of 70-500 bp,among which 314 were polymorphic,accounting for 81.89%of the total.The average values of observed allele number,effective allele number,Nei diversity indexand Shannon information index in 10 populations were 1.4573±0.0905,1.3101±0.0611,0.1702±0.0297 and 0.2514±0.0487,respectively.The genetic distance of the 10 populations ranged from 0.0454 to 0.1563,and the similarity coefficient ranged from 0.8234 to 0.9786.The dendrogram of Jingdong chestnuts was obtained by UPGMA clustering.The study of the genetic structure of the main export Jingdong chestnut populations in China aims to provide basic data for the rational assessment and protection of genetic resources,genetic breeding,and quality improvement,and maintain the international market reputation of chestnuts.
