摘 要:目的探讨人参-熟地黄水提物(RS-DH)对果蝇寿命的影响及潜在作用机制。方法将3日龄W118型雄性果蝇随机分为对照组和不同剂量(5、10 mg/mL)的人参-熟地黄水提物组,每组200只,记录果蝇的每日死亡数量和寿命;通过百草枯应激损伤模型和过氧化氢损伤模型,测定人参-熟地黄水提物对雄性果蝇存活时间的影响;通过试剂盒和q RT-PCR测定果蝇体内超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)活力和丙二醛(malondialdehyde,MDA)含量和与衰老相关基因的转录水平,探讨潜在的作用机制。结果寿命实验结果显示,与对照组果蝇相比,RS-DH 5 mg/mL组果蝇和RS-DH 10 mg/mL组果蝇均能延长果蝇的寿命,以5 mg/mL组延长最为显著(P<0.01)。百草枯应激损伤模型和过氧化氢损伤模型试验显示,与对照组相比,人参-熟地黄水提物可延长果蝇的存活时间。人参-熟地黄水提物能高体内抗氧化酶CAT和SOD的活性,降低MDA含量(P<0.05,P<0.01),显著上调SOD和CAT mRNA的表达和显著下调MTH mRNA的表达(P<0.05)。结论人参-熟地黄提取物可以延缓雄性果蝇寿命,其作用机制可能与提高果蝇抗氧化应激能力及调控长寿基因有关。Objective To investigate the effect of ginseng-radix rehmanniae praeparata(RS-DH)aqueous extract on the lifespan of drosophila melanogaster and its potential mechanisms.Methods Three-day-old male W118 wild type flies were randomly divided into a control group and different-dose(5 mg/mL,10 mg/mL)RS-DH aqueous extract groups,with 200 flies in each group.The daily death number and life span of the flies were recorded.The effect of RS-DH aqueous extract on the survival time of male drosophila was measured in the paraquat stress injury model and the hydrogen peroxide injury model.The activities of superoxide dismutase(SOD)and catalase(CAT),the content of malondialdehyde(MDA)and the transcription levels of aging-related genes in drosophila were measured by reagent kits and qRT-PCR,and its potential mechanisms of action were explored.Results Compared with the control group,the 5 mg/mL RS-DH group and the 10mg/mL RS-DH group extended the lifespan of drosophila,while the 5mg/mL RS-DH group was more significant(P<0.01).Compared with the control group,the experiments showed that RS-DH aqueous extract extended the survival time of flies in the paraquat stress injury model and the hydrogen peroxide injury model,increased the activities of CAT and SOD,and reduced the content of MDA(P<0.05,P<0.01),significantly up-regulated the SOD and CAT mRNA expression and significantly downregulated the MTH mRNA expression(P<0.05).Conclusion The RS-DH aqueous extract prolongs the lifespan of male drosophila,which may be related to improving the ability of anti-oxidative stress and regulating longevity genes in drosophila.
