摘 要:目的通过观察小鼠围着床期子宫内膜促血管生成因子1(angiopoietins 1,Ang1)、酪氨酸蛋白激酶2(tyrosine kinase receptors 2,Tie2)的表达,研究寿胎丸改善控制性超促排卵(controlled ovarian hyperstimulation,COH)后围着床期小鼠子宫内膜容受性的作用机制。方法选取未孕小鼠,随机分成正常组、模型组、寿胎丸低剂量组、寿胎丸中剂量组、寿胎丸高剂量组,分别予以醋酸亮丙瑞林微球、尿促性腺激素(human menopausal gonadotropin,HMG)、人绒毛膜促性腺激素(human chorionic gonadotophin,HCG)注射造模。0.9%氯化钠溶液、不同浓度中药灌胃。造模成功后,各组分别与雄性小鼠合笼,于合笼成功第2天(D2)、第4天(D4)、第6天(D6)取小鼠子宫内膜组织,运用免疫组化法及Western blot法检测Ang1、Tie2蛋白的表达情况。结果与模型组比较,寿胎丸中、高剂量组小鼠子宫内膜Ang1、Tie2表达量增加,差异具有统计学意义(P<0.05)。结论(1)COH减少围着床期小鼠子宫内膜Ang1、Tie2的表达,干扰子宫内膜血管生成,从而降低子宫内膜容受性。(2)寿胎丸可通过提高COH后小鼠围着床期Ang1、Tie2的表达,促进子宫内膜血管生成,改善子宫内膜容受性。Objective By observing the expressions of angiopoietins1(Ang1)and tyrosine kinase receptors 2(Tie2)in mice endometrium during peri-implantation period,to study the mechanism of Shoutai Pill(寿胎丸)improving the endometrial receptivity of mice in peri-implantation period after controlled ovarian hyperstimulation(COH).Methods Non-pregnant mice were selected and randomly divided into normal group,model group,Shoutai Pill low dose group,Shoutai Pill medium dose group and Shoutai Pill high dose group.They were given leuprorelin acetate microspheres,human menopausal gonadotropin(HMG),human choionic gonadotophin(HCG)injection in order to set up the model,and normal saline,different concentrations of traditional Chinese medicine were given by gavage.After successful modeling,each group was mixed up with male mice.Then uterine tissues were taken out on the 2nd,4 th and 6th day of the successful cage so as to detect the expressions of Ang1 and Tie2 proteins by technologies of immunohistochemistry method and Western blot.Results Compared with those of the model group,the expressions of Ang1 and Tie2 in the endometrium of mice increased in Shoutai Pill medium and high dose groups,and the difference was statistically significant(P<0.05).Conclusion①The expressions of Ang1 and Tie2 in mice endometrium during peri-implantation period were reduced by COH.It demonstrates that COH can affect endometrial receptivity by interfering with endometrial ngiogenesis.②Shoutai Pill increased the expressions of Ang1 and Tie2 in mice endometrium during peri-implantation period after COH.The effect of Shoutai Pill is beneficial to endometrial ngiogenesis and endometrial receptivity.
