摘 要:目的:研究佛甲草正丁醇提取物抗炎、镇痛作用,并初步探讨其机制。方法:采用二甲苯诱导的小鼠耳廓肿胀急性炎症模型、小鼠肉芽增生慢性炎症模型、完全弗氏佐剂诱导的大鼠关节炎模型、坐骨神经分支结扎所致的大鼠神经病理性疼痛模型、冰醋酸诱导的小鼠疼痛模型和热辐射痛模型,检测小鼠胸腺指数、脾脏指数、足趾肿胀度、耳廓肿胀率、热缩足反应、机械缩足反应、扭体次数、甩尾时间,酶联免疫学方法检测小鼠血清IL-10和IL-6含量,化学比色法检测SOD、GSH-Px和MDA含量。结果:佛甲草正丁醇提取物能有效减轻小鼠耳廓肿胀度、棉球所致的肉芽肿增生及佐剂性关节炎大鼠足趾肿胀度(P<0.05);明显提高神经病理性痛大鼠机械痛阈值(P<0.01),显著延长辐射热引起的小鼠疼痛甩尾反应潜伏期(P<0.001),抑制冰醋酸刺激小鼠腹腔黏膜引起的疼痛反应(P<0.01);下调小鼠脾及胸腺指数(P<0.01);降低小鼠血清IL-6和MDA含量(P<0.01),升高IL-10(P<0.01)、SOD及GSH-px含量(P<0.05)。结论:佛甲草正丁醇提取物具有抗炎镇痛作用,其作用机制可能与调节机体免疫功能、抑制炎症反应和抗氧化功能有关。Objective:To study the anti-inflammatory and analgesic effects of the Sedum lineare Thunb.n-butyl alcohol(SLNP),and preliminarily explore its anti-inflammatory mechanism.Methods:Six animal models were adopted,including Xylene-Induced ear edema in mice for acute inflammation,cotton pellet induced granuloma in mice for chronic inflammation,rats arthritis induced by Complete Freund's adjuvant,rats neuropathic pain model induced by the spared nerve injury,mice pain model induced by glacial acetic acid,and mice heat radiation pain model.Then we measured thymus index,spleen index,foot volume,the swelling rat of auricle,paw withdraw threshold(PWT),writhing times,tail flick latency,and meanwhile respectively detected the levels of IL-10 and IL-6 by enzyme-linked immunosorbent assay,and levels of SOD,GSH-Px and MDA by chemical colorimetry.Results:SLNP treatment inhibited the ear edema in mice caused by xylene,suppressed the proliferation of granuloma tissue(P<0.05),decreased the foot volume of arthritis rats,increased the pain threshold of rats subjected by SNI(P<0.01),significantly prolonged the tail flick latency of mice(P<0.001)and relieved the pain reaction caused by glacial acetic acid stimulation of abdominal mucosa(P<0.01),while decreased the thoracic gland index and spleen index(P<0.01).SLNP treatment also suppressed the IL-6 level(P<0.01)and promoted the IL-10 content in serum of mice(P<0.01).Otherwise,SLNP treatment inhibited the MDA content and improved activation of SOD and GSH-Px in serum of mice(P<0.05).Conclusion:SLNP has effects on analgesia and anti-inflammation,and the mechanism is probably related to the regulation of immunity,and inhibition of inflammation and antioxidant.
