摘 要:目的:探索莱菔子对正常高值血压痰湿壅盛证大鼠肝脏的保护作用及其机制。方法:以正常高值血压痰湿壅盛证大鼠为研究对象,以高脂饲料喂养及腹腔注射左旋硝基精氨酸(NG-Nitro-L-arginine,L-NNA)溶液(7.625 mg·kg^(-1))诱导正常高值血压痰湿壅盛证大鼠。符合纳入标准的模型大鼠随机分为模型组、莱菔子组与水飞蓟宾组。其中莱菔子组以莱菔子溶液(2.5 g·kg^(-1))灌胃,水飞蓟宾组以水飞蓟宾溶液(20 mg·kg^(-1))灌胃,空白组给予等质量生理盐水灌胃,每日1次。6周后,使用全自动生化分析仪对大鼠血清AST、ALT、TG、TC、LDL-C含量进行检测。HE染色观察肝脏组织形态和结构。Western Blot法检测大鼠肝脏组织chemerin、IL-6、CMKLR1、TNF-α、GAPDH、β-actin蛋白表达水平。ELISA法检测大鼠血清IL-6抗体、TNF-α水平。结果:模型组大鼠血清AST、ALT、TG、TC、LDL-C、TNF-α、IL-6水平明显高于空白组(P<0.05),莱菔子组血清AST、ALT、TG、TC、LDL-C、TNF-α、IL-6水平显著低于模型组(P<0.05)。模型组大鼠肝脏出现大量体积大小不一的脂滴空泡及炎性细胞浸润与细胞坏死。莱菔子组大鼠肝脏细胞排列整齐,脂肪空泡已经几乎消失,炎性细胞浸润略微减少,坏死细胞减少。模型组大鼠肝脏组织中的chemerin、CMKLR1、TNF-α、IL-6蛋白表达显著高于空白组(P<0.05),莱菔子组大鼠肝脏组织中的chemerin、CMKLR1、TNF-α、IL-6蛋白表达明显低于模型组(P<0.05)。结论:莱菔子对正常高值血压痰湿壅盛证大鼠肝脏具有明显的保护作用,降低chemerin与CMKLR1表达,抑制肝脏库普弗细胞(KCs)的激活,减少TNF-α与IL-6的释放可能是其机制之一。Objective:To explore the protective effect and mechanism of semen raphani on liver of rats with high-normal blood pressure of phlegm-dampness syndrome.Methods:The rats with high-normal blood pressure of phlegm-dampness syndrome were used as the research objects.In order to make such rat model,the rats were fed with high fat diet and injected with L-NNA solution(7.625 mg·kg^(-1)).Model rats meeting the inclusion criteria were randomly divided into model group,semen raphani group and silybin group.Semen raphani group was gavaged with semen raphani solution(2.5 g·kg^(-1)),silybin group was administered with silybin solution(20 mg·kg^(-1))by gavage,and blank group was gavaged with equal quality normal saline,once a day.After 6 weeks,the levels of TC,TG,LDL-C,AST and ALT in serum were measured using automatic biochemical analyzer.The morphology and structure of the liver tissue were observed through HE staining.The protein levels of chemerin,IL-6,CMKLR1,TNF-α,GAPDH andβ-actin in the rat liver were detected by Western Blot.Serum levels of IL-6 antibody and TNF-αwere determined through ELISA.Results:Serum levels of AST,ALT,TG,TC,LDL-C,TNF-αand IL-6 in model group were significantly higher than those in blank group(P<0.05).Serum levels of AST,ALT,TG,TC,LDL-C,TNF-αand IL-6 in semen raphani group were significantly lower than those in model group(P<0.05).A large number of lipid droplet vacuoles of varying sizes,inflammatory cell infiltration,and cell necrosis appeared in the liver of model group rats.The liver cells of rats in the semen raphani group were arranged neatly,fat vacuoles almost disappeared,inflammatory cell infiltration was slightly reduced,and necrotic cells were reduced.The protein expressions of chemerin,CMKLR1,TNF-αand IL-6 in the liver tissue of model group were significantly higher than those of blank group(P<0.05),and the protein expressions of chemerin,CMKLR1,TNF-αand IL-6 in the liver tissue of semen raphani group were significantly lower than those in model group(P<0.05).Conclusion:Semen ra
