通督调神灸对大脑中动脉阻塞大鼠学习记忆及轴突再生的影响  

Effects of Tongdu Tiaoshen Moxibustion on Learning and Memory Ability and Axonal Regeneration in Middle Cerebral Artery Occlusion Rats

摘　　要：目的:探究通督调神灸干预对大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)大鼠海马神经元凋亡、学习记忆及轴突再生的影响。方法:将SD大鼠45只随机分为3组:假手术组、模型组和通督调神灸组,15只/组。除假手术组外其他两组大鼠进行MCAO造模。造模后,对通督调神灸组取“百会穴”进行温和灸,20 min/次,1次/d,共7 d。采用Morris水迷宫观测大鼠学习记忆能力,micro-MRI扫描观察大鼠脑梗死体积,免疫组化和免疫荧光观察大鼠轴突再生标志性蛋白GAP-43表达情况,Western Blot检测Cofilin的表达。结果:与模型组相比,通督调神灸组和假手术组的水迷宫逃避潜伏期短,穿越平台次数增加,micro-MRI扫描结果发现通督调神灸组脑梗死体积百分比小于模型组,通督调神灸组的平均光密度明显高于模型组,模型组和通督调神灸组的红色荧光(GAP-43)强度均明显高于假手术组,通督调神灸组Cofilin表达量较模型组降低,差异均具有统计学意义(P<0.05)。结论:通督调神灸有利于减轻MCAO大鼠的神经功能缺损程度,减少脑梗死体积,抑制脑梗死区域神经元凋亡,同时抑制Cofilin表达,促进神经元轴突再生,促进学习记忆功能恢复。Objective:To detected the effecacy of Tongdu Tiaoshen moxibustion on neuronal apoptosis,ability of learning and memory and axon regeneration in rats with middle cerebral artery occlusion(MCAO).Methods:45 rats were randomly divided into three group:sham operation group,MCAO model group and Tongdu Tiaoshen moxibustion(MOX)group,15 rats in each group.Except sham operation group,the other rats were made in MCAO.MOX group was intervented with moxibustion on“Baihui”for 20 minutes,once daily,lasted for 7 days.Detected ability of learning and memory of rats with Morris water maze,and cerebral infarct,neuronal apoptosis,axon regeneration and expression of Cofilin were detected by micro-MRI,TUNEL,immunohistochemistry and immunofluorescence staining of GAP-43 and Western Blot.Results:Compared with the model group,the escape latency of MOX group and the sham operation group were shortened,the time of crossing the platform was more frequent in Morris test,the cerebral infarction volume was smaller,the average optical density of the MOX group was significantly higher,the expression level of Cofilin protein was decreased,all<0.05.Conclusion:After treatment with Tongdutiaoshen moxibustion for 7 days,the neurological impairment was improved,the cerebral infarction volume of MCAO rats decreased,and at the same time inhibit the expression of Cofilin,promote the regeneration of axons,then improved the learning and memory of MCAO rats.
