摘 要:为探究急性高温胁迫对黑龙江茴鱼(Thymallus arcticus grubei)生理适应性的影响,采用生物化学和分子生物学的方法研究了急性高温下(11、14、17℃和20℃)黑龙江茴鱼血清生化指标、消化酶和抗氧化酶等变化规律。结果显示:急性高温胁迫过程中,黑龙江茴鱼血清生化指标发生显著变化。其中,17℃组和20℃组血清中谷草转氨酶(AST)和谷丙转氨酶(ALT)活性随胁迫时间的增加显著升高,血清中总蛋白(TP)含量和碱性磷酸酶(ALP)活性则随胁迫时间的延长呈下降趋势。黑龙江茴鱼肠道淀粉酶和蛋白酶活性随高温胁迫时间的增加呈先上升后下降的变化趋势。在高温胁迫24 h时,17℃组和20℃组黑龙江茴鱼肠道的淀粉酶、蛋白酶和脂肪酶活性均显著高于对照组;于胁迫48 h时,其肠道淀粉酶和蛋白酶活性显著降低,而脂肪酶活性显著增加。此外,黑龙江茴鱼肝脏中的超氧化物歧化酶(SOD)和谷胱甘肽抗氧化物酶(GPX)活性随高温胁迫时间的延长显著升高,而过氧化氢酶(CAT)活性则在高温胁迫48 h时显著下降。肝脏中SOD和CAT基因相对表达量在急性高温胁迫下显著上调表达。研究结果表明,在急性高温处理下黑龙江茴鱼在17~20℃下处于胁迫状态,其血清生化指标、消化酶、抗氧化酶活性及其基因表达受到显著影响。Biochemical indexes,digestive enzymes and antioxidant enzymes in the serum of the fish under acute high temperature(11℃,14℃,17℃and 20℃)were studied by biochemical and molecular biological methods to explore the effect of acute high-temperature stress on the physiological adaptability of Thymallus arcticus grubei.The results showed that the serum biochemical indexes of T.grubei changed significantly under acute high-temperature stress.Among them,the activities of glutamic oxalate transaminase(ALT)and alanine transaminase(AST)in serum of 17℃and 20℃groups increased significantly with the increase of stress time.In contrast,the content of total protein(TP)and the activity of alkaline phosphatase(ALP)in serum decreased with the increase of stress time(P<0.05).The activities of amylase and protease in the intestine of T.grubei increased first and then decreased with the increase of high-temperature stress time.The activities of amylase,protease and lipase in the intestine of T.grubei in 17℃and 20℃groups were significantly higher than those in the control group at 24 hours of high-temperature stress.At 48h of stress,the activities of amylase and protease in the intestine decreased significantly,while the activities of lipase increased significantly.In addition,the activities of superoxide dismutase(SOD)and glutathione antioxidant enzyme(GPX)in the liver of T.grubei increased significantly with the extension of heat stress time,while the activity of catalase(CAT)decreased significantly at 48 h of heat stress.The liver’s relative expression of SOD and CAT genes was significantly up-regulated under acute heat stress.The results showed that under acute high-temperature treatment,T.grubei was under stress at 17~20℃,and its serum biochemical indexes,digestive enzymes,antioxidant enzyme activities and gene expression were significantly affected.Therefore,during the production and operation of T.grubei,we should pay close attention to the change in culture temperature and try to avoid the stress damage cause
