摘  要:在北方寒冷地区,冷应激不可避免的带来一系列负面影响。试验探讨在冷暴露环境下添加不同浓度的盐酸川芎嗪对小鼠肝脏产热能力的影响。冷暴露前对预饲一周后的小鼠进行相应的灌胃处理,常温对照组每天始终保持在26±2℃环境中饲养,冷暴露组每天置于4℃人工气候室中冷刺激3 h。四周后采集小鼠的肝脏组织,采用HE染色法观察肝脏组织是否有损伤;采用油红O染色法观察肝脏组织中脂滴的分布和数量;采用IHC法和Western Blot法对肝脏组织中解偶联蛋白1(UCP1)的表达量进行检测。与对照组相比,添加了盐酸川芎嗪调后肝脏无损伤,肝脏中脂滴数增加,产热因子UCP1的表达量显著提高。研究结果表明,冷暴露下盐酸川芎嗪可提高UCP1的表达量,促进小鼠肝脏产热,为缓解冷应激产生的负面影响提供一定的参考。In cold northern regions,cold stress inevitably brought a series of negative effects.The effects of adding different concentrations of chuanxiongzine hydrochloride on the liver thermogenic capacity of mice under cold exposure environment were investigated.The mice were gavaged accordingly after one week of pre-feeding before cold exposure,and the normothermic control group was kept at 26±2℃every day,while the cold-exposed group was cold-stimulated in an artificial climate chamber at 4℃for 3 h.Liver tissues of mice were collected after four weeks,and HE staining was used to observe whether there was any damage to liver tissues;oil red O staining was used to observe the distribution and number of lipid droplets in liver tissues;IHC method and Western Blot method were used to detect the expression of uncoupling protein 1(UCP1)in liver tissues.Compared with the control group,there was no damage to the liver after the addition of Ligustrazine hydrochloride,the number of lipid droplets in the liver increased,and the expression of the thermogenic factor UCP1 was significantly higher.The results showed that,ligustrazine hydrochloride under cold exposure could increase the expression of UCP1 and promote heat production in the liver of mice,providing some reference for alleviating the negative effects produced by cold stress.
