一例奶牛气肿疽猝死病例的实验室诊断和分析

摘  要:为给某奶牛养殖户多头奶牛突然死亡的原因作出科学判定,对病死奶牛实施流行病学调查、临床观察、尸体剖检和微生物学及分子生物学实验室诊断等一系列检查。结果发现死亡奶牛出现急性败血症病理变化;经细菌分离培养,所采取的微生物学病料在血琼脂培养基厌氧培养24 h后,可见透明的、具有β溶血的革兰氏染色阳性菌落,在光学显微镜下呈现杆状;所分离细菌经聚合酶链式反应(PCR)检测,合成扩增细菌16S rDNA的通用引物扩增的DNA片段约1.6 kb左右,细菌DNA片段与气肿疽梭菌相符。经流行病学调查、临床症状观察、尸体剖检、微生物培养鉴定和分子生物学实验室检测等,确诊为奶牛气肿疽病,为奶牛气肿疽病的临床防控提供了实验依据和理论建议,可有效降低气肿疽病给奶牛养殖造成的经济损失及对公共卫生安全造成的威胁。In order to make a scientific judgment on the cause of the sudden death of many dairy cows in a dairy farmer,a series of examinations such as epidemiological investigation,clinical observation,autopsy,and laboratory diagnosis of microbiology and molecular biology were carried out on the dead cows.The results showed that the dead cows had pathological changes of acute sepsis;after bacterial isolation and culture,the collected microbiological disease materials were cultured in blood agar medium anaerobic for 24 hours,and transparent Gram-positive colonies withβ-hemolysis could be seen.Under the light microscope,it is rod-shaped;the isolated bacteria were detected by polymerase chain reaction(PCR),and the DNA fragment amplified by the universal primer for synthesizing and amplifying bacterial 16S rDNA was about 1.6 kb,and the bacterial DNA fragment was consistent with Clostridium anthracis.After epidemiological investigation,clinical symptom observation,necropsy,microbial culture identification and molecular biology laboratory test,the diagnosis of cow emphysema anthracnose was made.It provided experimental basis and theoretical suggestions for the clinical prevention and control of cow emphysema,which could effectively reduce the economic loss caused by emphysema to dairy cattle breeding and the threat to public health security.
