摘 要:以番茄砧木种子为材料,以蛭石为引发基质,研究了不同温度引发对种子发芽特性、幼苗生长及耐冷性的影响,以筛选出适合番茄砧木种子引发的温度。结果表明,与未进行引发处理的CK相比,10、15、25、5℃/20℃种子引发可不同程度地促进种子萌发、增大壮苗指数,降低幼苗低温逆境下的冷害指数和电解质渗漏率,并通过增加可溶性糖和可溶性蛋白质含量,提高超氧化物歧化酶、过氧化物酶和过氧化氢酶活性及根系活力增强耐冷性;低温10、15℃和变温5℃/20℃引发效果优于常温25℃引发,其中5℃/20℃引发的发芽率、发芽指数、壮苗指数分别是CK的1.58倍、5.90倍和2.43倍,低温胁迫下可溶性糖和可溶性蛋白质含量分别是CK的4.0倍和1.5倍,且冷害指数最低。综上及耐冷指标的隶属函数值可知,变温5℃/20℃引发效果最好,其次为10℃和15℃恒温引发。To screen out the suitable seed priming temperature,we studied the effects of different priming temperature on seed germination,seedling growth and cold resistance of tomato rootstock using vermiculite as the priming substrate.The results showed that compared with CK without priming treatment,seed priming at 10℃,15℃,25℃and 5℃/20℃could promote seed germination,increase seedling strength index,reduce chilling injury index and electrolyte leakage of seedlings under chilling stress,and improve cold tolerance by increasing soluble sugar and soluble protein content,the activities of superoxide dismutase,peroxidase and catalase,and root activity.The priming effect at low temperatures of 10℃,15℃and variable temperatures of 5℃/20℃were better than that at room temperature of 25℃.Among them,the germination rate,germination index and seedlings strength index after priming at 5℃/20℃were 1.58 times,5.90 times and 2.43 times that of CK,respectively.Under low temperature stress,the soluble sugar and soluble protein content were 4.0 times and 1.5 times that of CK,respectively,and the cold damage index of 5℃/20℃treatment was the lowest.According to the above indexes and the membership function values of the cold resistance,it can be seen that the priming effect at changing temperature of 5℃/20℃was the best,followed by the constant temperature priming at 10℃and 15℃.
