摘 要:目的:探讨肉桂油对Ⅱ型糖尿病心肌病模型大鼠心肌的保护作用。方法:SD大鼠168只,分为正常对照组(20只)和实验组(148只)。正常对照组不做任何处理,实验组建立高脂高糖联合链脲佐菌素(STZ)糖尿病模型,剔除未达糖尿病标准的大鼠,将剩余实验组大鼠随机分为模型对照组(N-ISO组、ISO组)、肉桂油小、中、大剂量组、胰岛素组,除正常对照组、N-ISO组外,各组再给予异丙肾上腺素(ISO),5 mg/kg,1 time/d,连续注射5天,各组同一时间开始给予相应药物,正常对照组给予等量蒸馏水,末次注射ISO 4周后进行指标检测。检测指标包括:大鼠心功能测定、大鼠心重指数测定、病理学检查。结果:与模型对照组(N-ISO组、ISO组)相比,肉桂油中、大剂量组左心室内压最大下降速率-dP/dtmax有所提高(P≤0.05);肉桂油中剂量、大剂量组与ISO组比较,其心重指数减小(P≤0.05);病理学显示,肉桂油大剂量组HE染色纤维化改变较ISO组程度轻,Masson胶原染色面积较ISO组明显较小(P≤0.05)。结论:肉桂油对糖尿病心肌损伤大鼠心功能改善具有一定的作用,能对抗糖尿病心肌病所引起的左心室肥大,并减少心肌胶原形成。Objective:This study aimed to investigate the protective effects of cinnamon oil on myocardial damage in a type II diabetic cardiomyopathy rat model.Methods:A total of 168 Sprague-Dawley rats were divided into a normal control group(20 rats)and an experimental group(148 rats).The normal control group underwent no treatment,while the experimental group was induced with a high-fat,high-sugar diet in combination with streptozotocin(STZ)to establish a diabetes model.Rats that did not meet the diabetes criteria were excluded,and the remaining rats in the experimental group were randomLy divided into model control,low,medium,and high-dose cinnamon oil groups,as well as an insulin group.All groups(except the normal control group)were administered with isoproterenol(ISO)at 5 mg/kg,once daily,for 5 consecutive days.The corresponding medications were started simultaneously in each group,with the normal control group receiving an equivalent amount of distilled water.The indicators were assessed 4 weeks after the final ISO injection,including rat cardiac function,heart weight index,and histological examination.Results:Compared to the model group(ISO group and N-ISO group),the medium and high-dose cinnamon oil groups showed an improved left ventricular maximal rate of pressure decline(dP/dtmax)(P≤0.05).The medium and high-dose cinnamon oil groups exhibited an increased T wave amplitude(P≤0.05)and a decreased R wave amplitude(P≤0.05)in animal electrocardiograms.The high-dose cinnamon oil group demonstrated a reduced heart weight index compared to the model group.Remarkably,the high-dose cinnamon oil group displayed a smaller area of collagen staining in Masson's trichrome staining(P≤0.05)compared to the model group.Conclusion:Cinnamon oil has a certain role in improving cardiac function in diabetic rats with myocardial injury and can counteract left ventricular hypertrophy caused by diabetic cardiomyopathy while reducing myocardial collagen formation.
