摘  要:目的:建立地黄药材、地黄饮片中寡糖类成分的薄层色谱鉴别方法。方法:以水苏糖和地黄对照药材为对照品,对供试品溶液的制备方法、展开剂(乙酸乙酯-乙醇-水-乙酸,乙酸乙酯-乙醇-水-氨水及其不同配比)、显色检视方法(日光和紫外366nm)进行薄层色谱鉴别方法的考察优化,并对温度、湿度、不同品牌预制板分别进行了方法耐用性考察,确定地黄的供试品溶液制备方法和最佳薄层色谱条件。结果:采用硅胶G板,以乙酸乙酯-乙醇-水-氨水(2∶5∶4∶0.5)为展开剂,喷以10%硫酸乙醇溶液,在105℃加热至斑点显色清晰。地黄药材、地黄饮片供试品色谱中,在与对照药材色谱相应的位置上,显相同颜色的斑点,阴性对照相同位置无干扰。结论:新建方法简便快捷、专属性强、灵敏度高,可同时应用于地黄药材和地黄饮片,为地黄提供了快速定性鉴别方法和依据。Objective:To establish a TLC method for the identification of oligosaccharides in radix Rehmannia and radix Rehmannia decoction pieces.Methods:Using stachyose and Rehmannia glutinosa as reference materials,the preparation method of the test solution,the developing agent(ethyl acetate ethanol water acetic acid,ethyl acetate ethanol water ammonia and their different ratios),and the colorimetric examination method(sunlight and UV 366 nm)were optimized for thin layer chromatography identification.The durability of the method was also investigated for temperature,humidity,and different brands of prefabricated boards,Determine the preparation method and optimal thin-layer chromatography conditions for the test solution of Rehmannia glutinosa.Results:Using a silica gel G plate,using ethyl acetate ethanol water ammonia(2∶5∶4∶0.5)as the developing agent,spray a 10%sulfuric acid ethanol solution,and heat at 105℃until the spots show clear color.In the chromatogram of Rehmannia officinalis and Rehmannia decoction pieces,spots of the same color appear at the corresponding positions of the reference medicinal material chromatogram,and there is no interference at the same position of the negative control.Conclusion:The new method is simple,fast,specific,and highly sensitive,and can be applied to both Rehmannia glutinosa medicinal materials and Rehmannia decoction pieces,providing a rapid qualitative identification method and basis for Rehmannia glutinosa.
