摘 要:本文设计了一个合成细胞内溶酶体定位的NO荧光探针并进行细胞成像的本科药物化学实验。首先,将可对NO响应的荧光物质进行结构修饰,通过一定的连接方式将溶酶体定位基团引入荧光物质中,合成了新型NO荧光探针;其次,对所合成的探针进行简单的体外性能测试,通过对NO溶液滴定的荧光、吸收光谱的分析以及对不同pH值和不同生物分子的荧光响应情况的评估,揭示了该探针的高灵敏度、高选择性;最后,通过细胞成像实验,证实了探针能选择性检测溶酶体内的内源性的NO。将该实验引入到药物化学实验教学中,可以提升学生的实验操作能力,了解当前药物化学实验的前沿研究方向,增强学生的科研素养,为其将来更好地从事药物化学相关的研究打下基础。A medicinal chemistry experiment for undergraduates was designed by synthesizing and bioimaging of a novel fluorescent NO probe targeting intracellular lysosome.Firstly,a novel NO fluorescent probe was synthesized by linking a lysosome targeting group to a fluorescent material that specifically response to NO.Secondly,a series of in vitro test of the probe were performed including NO titration,pH conditions impact,and possible interferences of many other biologically relevant species,suggesting high sensitivity,pH stability,and selectivity.Finally cellular imaging assays were conducted to verify the probe’s ability to track endogenous NO in lysosomes in living cells.The introduction of this experiments to the medicinal chemistry experiments teaching can enhance the undergraduates’operation ability and academic ability,understand the frontier research area in medicinal chemistry,which will eventually be helpful to their future academic research related to medicinal chemistry.
