摘  要:目的采用含有钆元素的化合物及羟基磷灰石(HA)为原料,通过化学合成法制备一种新型纳米含钆化合物的羟基磷灰石复合支架,为组织工程中软骨损伤修复提供新型工程支架。方法采用均相沉淀法进行制备含钆化合物的羟基磷灰石复合支架,并自备成圆柱形立体结构,对含钆化合物的羟基磷灰石复合支架进行外观大体形态及空隙率观察评估,进一步在电镜下观察孔隙率、形态结构是否满足实验要求。结果制备的含钆化合物的羟基磷灰石复合支架,外观形态良好,空隙率较大,大孔结构有利于骨组织生长;电镜下扫描支架内可见孔隙为蜂窝状结构,相互连通性良好,行表面观察见羟基磷灰石颗粒呈不规则多边形,相互紧密排列,但个别颗粒之间有微孔形成。如此大孔隙与微孔隙共存的三维立体结构,为种子细胞的增殖、分化提供了良好的支架条件。结论本实验制备的含钆化合物的羟基磷灰石复合支架材料,其外观及电镜下已经具有很好的孔隙率及大孔径的三维立体微孔结构,但是其支架的细胞毒性及共培养后细胞增殖能力的影响,仍需要进一步深入研究。Objective The compound containing gadolinium and hydroxyapatite(HA)were used as raw materials,a novel nano-hydroxyapatite composite scaff old containing gadolinium compound was prepared by chemical synthesis,to provide a new engineering scaff old for cartilage repair in tissue engineering.Methods The hydroxyapatite composite scaff old containing gadolinium compounds was prepared by homogeneous precipitation method,and the cylindrical three-dimensional structure was prepared.The appearance and general morphology of the hydroxyapatite composite scaffold containing gadolinium compounds were evaluated for observation and evaluation,and the porosity and morphological structure were further observed under electron microscopy to see whether the porosity and morphological structure met the experimental requirements.Results The prepared hydroxyapatite composite scaff old containing gadolinium compounds has good appearance,large porosity and large pore structure,which is conducive to bone tissue growth.Under electron microscopy,the pores in the scaffold were honeycomb structures with good mutual connectivity.The hydroxyapatite particles were irregular polygons closely arranged with each other,but micropores were formed between individual particles.The three-dimensional structure of such large pores and micro-pores provides a good scaff old for the proliferation and diff erentiation of seed cells.Conclusion The hydroxyapatite composite scaff old containing gadolinium compounds prepared in the experiment has a good porosity and a three-dimensional micropore structure with large pore size under the appearance and electron microscopy,but the cytotoxicity of the scaff old and the eff ects of co-culture on cell proliferation still need further study.
